This episode reviews the evidence for the prenatal estrogen theory of being transgender in natal males. Since estrogen does not readily penetrate the brain, the evidence cited for the estrogen theory involves the synthetic chemical diethylstibestrol or DES. This drug was given to pregnant females during 1940's through the 1980's but was eventually banned because it caused cancer in female offspring who were exposed through their mothers in utero. Because of this, most of the research was conducted with exposed natal females. Research with exposed males was initiated by DES-esposed researchers who believed that they were affected to become transgender. They started a website for DES exposed males and found that 150 of the 500 males who joined the website thought that they were transgender. This episode provides an answer as whether the evidence supports whether DES causes being transgender and the status of the estrogen theory of being transgender.

In higher organisms, all cells share the same genome, but every cell expresses only a limited and specific set of genes that defines the cell type. During cell division, not only the genome, but also the cell type is inherited by the daughter cells. This intriguing phenomenon is achieved by a variety of processes that have been collectively termed epigenetics: the stable and inheritable changes in gene expression patterns. This article reviews the extremely rich and exquisitely multi-scale physical mechanisms that govern the biological processes behind the initiation, spreading and inheritance of epigenetic states. These include not only the changes in the molecular properties associated with the chemical modifications of DNA and histone proteins, such as methylation and acetylation, but also less conventional ones, such as the physics that governs the three-dimensional organization of the genome in cell nuclei. Strikingly, to achieve stability and heritability of epigenetic states, cells take advantage of many different physical principles, such as the universal behavior of polymers and copolymers, the general features of non-equilibrium dynamical systems, and the electrostatic and mechanical properties related to chemical modifications of DNA and histones. By putting the complex biological literature under this new light, the emerging picture is that a limited set of general physical rules play a key role in initiating, shaping and transmitting this crucial "epigenetic landscape". This new perspective not only allows to rationalize the normal cellular functions, but also helps to understand the emergence of pathological states, in which the epigenetic landscape becomes dysfunctional.

Prenatal Testosterone Theory of Transgender (PTTT) Causation: Origins and Evidence. Do low prenatal testosterone levels in males and do high prenatal testosterone levels in females cause being transgender? The origins of this theory in eugenics and the evidence refuting the theory. The PTTT is rooted in eugenics theory developed in the US during the first half of the 20th Century. With support from private foundations, eugenics developed and spread to Germany and Europe. After World War II, East Germant eugenic endocrinologists developed the PTTT as the way to get rid of transgender people in society by adminstering drugs to pregnant mothers. If a mother was carrying a male baby, testosterone booster shots were recommended; if a mother was carrying a female baby, testosterone blockers were advised. The scientific evidence refutes the PTTT.

Our genes turn on and off and regulate our health and their illnesses. What we're telling them is that they're victims. If you don't like the traits, you can't change the genes, and the genes are turning on and off without you being involved. But Bruce left the university because his research revealed that the environment was controlling the genes. If the genes control you, then you're a victim of your heredity. If the environment is influencing your genes, then you can be a master because you're the one that can adjust to the environment. Epigenetics The idea of the nucleus controlling biology is because the nucleus contains 98% of the genes. But Bruce's research said, no, the nucleus is not the brain cell. In the sixties, he was doing experiments removing the nuclei of cells. It's called enucleation. What's interesting is if the nucleus was the brain of the cell, then by definition, if you remove the brain and throw it away, the organisms will die. And yet his cells lived for months with no genes in them. Removing a nucleus and throwing away the genes did not stop the cell from living or behaving in a very complex manner, which by definition says life was not an accident. Life is a lot of controlled behavior. If there are no genes, then what was controlling behavior? Biologists considered the cell membrane to be nothing more than plastic wrap to hold the cell together. And yet Bruce's research revealed that that simplicity was elegance, that simplicity was the basis of an information processor. The cell membrane is a carbon-based information processor. It reads environmental information and translates it into biological behavior. So it's an interface between the outside and the inside the membrane, the skin. Bruce's research revealed that it was controlling biology. He followed through, and 20 years later, science recognized the new field called epigenetics, which is the science Bruce described without having a name for it. Subconscious Mind You are the creator of this life. You are the one that creates wellness or disease. A gene does not cause cancer. Cancer is caused by lifestyle and environment, perceptions, beliefs. That's why spontaneous remissions of cancer are so significant because they reveal when a person changes their views, a disease can instantly disappear. And suddenly, you start to realize, oh my God, I am controlling this. I am the master of my biology. And once you understand that, freedom. Freedom from fear, fear of illness, disease, and sickness is just a reflection of weakness, not a body, but a deficiency of consciousness. Our thoughts and beliefs can alter every gene in our body to create over 3000 variations of every gene that we have in our body, which means what? We look at the world and respond to the world by adjusting our genetics. Less than 1% of disease is connected to genetics. 90% or more of disease is actually due to a lifestyle and belief and behavior that we're beginning to recognize that illness is not the result of organic foundation. Abstract Recent advances have begun to elucidate how epigenetic regulatory mechanisms are responsible for establishing and maintaining cell identity during development and adult life and how the disruption of these processes is, not surprisingly, one of the hallmarks of cancer. In this review, we describe the major epigenetic mechanisms (i.e., DNA methylation, histone and chromatin modification, non-coding RNA deployment, RNA editing, and nuclear reorganization) and discuss the broad spectrum of epigenetic alterations that have been uncovered in pediatric and adult nervous system tumors. We also highlight emerging evidence that suggests epigenetic deregulation is a characteristic feature of so-called cancer stem cells (CSCs), which are thought to be present in a range of nervous system tumors and responsible for tumor maintenance, progression, treatment resistance, and recurrence. We believe that better understanding how epigenetic mechanisms operate in neural cells and identifying the etiologies and consequences of epigenetic deregulation in tumor cells and CSCs, in particular, are likely to promote the development of enhanced molecular diagnostics and more targeted and effective therapeutic agents for treating recalcitrant nervous system tumors.

The carcinogenic effects of low dose radiation are not clear. Moreover, it is not known if exposure to low dose radiation increases cancer risk in offspring of irradiated parents. Radiation is known to damage DNA and cause mutations. Cancer arises in part through mutations in oncogenes and tumor suppressor genes. However, epigenetics is also known to play an important and perhaps central role in cancer induction. Epigenetics is here defined as a heritable change in phenotype without an underlying mutational event. Epigenetic alterations may be induced early in development as a result of diet or environmental exposure. These alterations can persist into adulthood, and may increase the risk of developing diseases such as cancer later in life. Radiation has been shown to cause epigenetic effects in mammalian cells and in mice, although the role of these in human cancer is entirely unknown. Here we hypothesize that the carcinogenic effect of radiation may in part be due to epigenetic alterations. In this study we will determine if early exposure to radiation leads to a change in DNA methylation in adult tissues and in ensuing unexposed generations, and if these changes correlate with increased cancer susceptibility. We also expect to identify a radiation exposure signature which when validated, could be used clinically as a biomarker of radiation exposure. These experiments will also open up new avenues of investigation into the role of epigenetics in cancer caused by radiation and other environmental carcinogens.

Our goals for this project are to explore LSD1 inhibition of protein-protein interactions as a potential therapy for ER- breast cancer, ameliorating iLCC for in vivo use, and using novel proteomics approaches to identify coregulatory proteins interacting with ER and LSD1 in ER+ cells and deduce how the complement of LSD1 associating proteins change in ER- cancers. We have made significant progress on the aims this project,specifically by examining the effects of small molecule, LSD1-selective and broad-spectrum histone demethylase inhibitors on breast cancer tumor growth as compared to iLCC. Using protein engineering, and H/D exchange MS,we identified the LSD1 alpha helical coiled-coil as a central mediator of ER signaling and coregulatory protein recruitment. Using siRNA knockdown and a newly identified highly selective inhibitor of the LSD1 demethylase active site chemistry, we showed that loss of LSD1 or inhibition of the LSD1-CoREST complex by iLCC inhibits breast cancer proliferation, but the antiproliferative effects of tranylcypromine or pargyline exhibit antiproliferativeeffects in ER+ and ER- breast cancers by a secondary mechanism requiring LSD1 as a scaffolding element.

Looping, the practice of keeping a group of children with the same teacher for more than year, has the potential to provide that consistency that is critical for attachment. The field of epigenetics and its compelling findings concerning attachment demand a second look at looping and how it can be implemented to maintain attachment, which is critical to the child's physical, cognitive, and emotional health now and in the future. This case study, conducted at a preschool that practices looping, examines the benefits and challenges of looping through the lens of applied epigenetics.

Intrauterine hyperglycemia is not only associated with increased perinatal morbidity and mortality, but also with increased lifelong risks of the exposed offspring for obesity, metabolic, cardiovascular and malignant diseases. Accumulating evidence suggests that fetal overnutrition (and similarly undernutrition) lead to persistent epigenetic changes in developmentally important genes, influencing neuroendocrine functions, energy homeostasis and metabolism. The concept of fetal programming has important implications for reproductive medicine. Because during early development the epigenome is much more vulnerable to environmental cues than later in life, avoiding adverse environmental factors in the periconceptional and intrauterine period may be much more important for the prevention of adult disease than any (i.e. dietetic) measures in infants and adults. A successful pregnancy should not primarily be defined by the outcome at birth but also by the health status in later life.

Abstract Females of the SWR/Bm (SWR) inbred mouse strain possess a unique susceptibility to juvenile-onset tumors originating from the granulosa cells (GC) of the ovarian follicles. Tumor susceptibility is an inherited, polygenic trait in SWR females, minimally involving an oncogenic Granulosa cell tumor susceptibility (Gct) locus on chromosome (Chr) 4 (Gct1), and two GC tumor susceptibility modifier genes mapped to distinct regions of Chr X (Gct4 and Gct6). Shifts in the frequency of GC tumor initiation in the SWR female population from low penetrance to moderate penetrance, or phenotype switching between GC tumor-susceptible and GC tumor-resistant, is strongly influenced by the allelic contributions at Gct4 and Gct6. In addition to the allele-specific effects, GC tumor susceptibility is controlled by the mode of X-linked transmission with a dominant, paternal parent-of-origin effect. We took advantage of the robust paternal effect with a recombinant male progeny testing strategy to resolve the Gct4 locus interval to 1.345 million base (Mb) pairs. Based on the mapping resolution and the phenotype sensitivity to endogenous and exogenous androgen exposure, a promising candidate for Gct4 identity is the androgen receptor (Ar) gene. We explored the mechanism of allelic variation for Ar between SWR (low penetrance allele) and SJL/Bm (SJL) (moderate penetrance allele) using an SWR.SJL-X congenic strain resource and a quantitative gene expression method. We report the low GC tumor penetrance allele of the SWR strain correlates with significantly reduced Ar transcript levels in the female ovary at the pubertal transition.

An understanding of the cancer cell begins with knowledge of the genetic alterations that lead to neoplastic transformation. Much progress has been made in identifying areas of deletion, amplification and mutation in tumors. However, this is only part of the picture. Increasingly, we are learning that epigenetic changes, that is, changes in chromatin structure, are critically important in regulating cellular gene expression. Recently, several labs have published manuscripts identifying RNA interference as being crucial for the establishment of such epigenetic changes in species as diverse as Drosophila, plants, and the fission yeast S. pombe. This knowledge presented a fantastic opportunity not only to study epigenetic changes, but to potentially create epigenetic changes by creating germline transgenic mice in which a target gene has been silenced by RNAi. After the demonstration that RNAi mammalian cells can be mediated by vectors encoding short, hairpin RNAs (shRNAs), we sought to develop a system by which to create transgenic mice using this technology. We demonstrate that a stable, heritable RNAi trigger in the form of a short hairpin was successfully passed through the mouse germline. As well as demonstrating a technique that can be applied to any gene of interest, we have created gene knock-down mouse models for the Neil-1 DNA glycosylase involved in DNA repair pathways, and for p53. These observations open the way to the use of RNAi as a complement to standard knockout methodologies and provides a means to rapidly assess the consequences of suppressing a gene of interest in a living animal. The vision driving the creation of this technology was one of eventual RNAi-based therapeutics. One could particularly imagine silencing oncogenes in cancer cells to attenuate their tumorigenic tendencies.

The objectives of this research are (1) to conduct a literature review to understand the extent and evidence on the influence of biodiversity exposure on epigenetic markers (CpG) and epigenetic clocks and (2) to develop a conceptual framework capturing the link between biodiversity and human health through epigenetics. Regarding the primary analysis of epigenetic variations as mediators, we will examine three categories of epigenetic changes associated with biodiversity exposure: (1) the biomarkers individual (CpGs), (2) the epigenetic clocks and pace of aging, (3) other epigenetic changes (histone modifications, non-coding RNAs). Each category will be evaluated based on the strength of evidence from existing studies. While epigenetic clocks are already established predictors of age-related outcomes, individual biomarkers and other modifications will be analysed for their potential mediation role in health effects. This study aims to advance our understanding of the links between biodiversity exposure, epigenetics modifications, and human health outcomes through the development of a conceptual framework. The framework could serve as a basis for further studies on this topic. The current evidence is limited to observational data, this emerging field holds promise for enhancing our knowledge of how environmental biodiversity impacts human health and well-being, providing a foundation for future investigations.
