Abstract Background: Angiogenesis, which is required for embryonic development and many physiological events, plays crucial role in many pathological conditions such as tumor growth and metastasis. Recent studies indicate anticancer and antitumor properties of saffron against human cancers. Many processes are affected by Electromagnetic Field (EMF) and its effect on proliferation and gene expression were examined. In this experimental study, the synergic effects of saffron and EMF on VEGFR2 gene expression in MCF7 cells were investigated. Methods: Saffron was extracted using freeze dryer. MCF7 cells were grown in RPMI 1640 medium supplemented with 10% FBS and incubated at 37°C with 5% CO2. After 24 hr cells were treated with saffron extract at concentrations of 100, 200, 400 and 800 µg/ml. Forty eight hr after treatment all flasks were exposed with EMF (50 Hz, 0.004 T). Then total RNA was extracted and cDNA was synthetized using specific primer. Synthetized products were analyzed by Real Time PCR to determine expression level of VEGFR2. Data were analyzed by SPSS (ANOVA & Tukey). Results: Critical inhibitory effect on VEGFR2 gene expression was 20% at 400 µg/ml. Synergic use of EMF and saffron extract showed most reduction (38%) at 100 µg/ml. On the other hand synergic use of 200, 400 and 800 µg/ml saffron aqua extract and EMF decline noticeably the VEGFR2 level of gene expression to 29, 35 and 36%, respectively. EMF itself also reduced VEGFR2 up to 25% in comparison with control group which is remarkable at p < 0.001. Conclusion: Results indicate a decrease in the expression of vascular endothelial growth factor receptor in the treated samples with saffron extract compared to control. This reduction in VEGFR2 level induced by synergic treatment of saffron and EMF which reveals induction of inhibitory effects of saffron on angiogenesis and could be also considered as a promising chemotherapeutic agent in breast cancer treatment.

Abstract Previous studies have indicated relaxant, inhibitory effect on histamine (H1) and muscarinic receptors, and stimulatory effect on β-drenoceptor of Crocus sativus on guinea pig tracheal chains. In the present study, the effect of the extract of C. sativus and one of its constituents, safranal, on the inflammatory changes of sensitized guinea pigs was examined. Eight groups of sensitized guinea pigs to ovalbumin were studied. One group was given drinking water alone (group S), while other 7 groups were received drinking water containing; three concentrations of safranal (4, 8 and 16 μg/ml), three concentrations of extract (0.1, 0.2 and 0.4 mg/ml) and one concentration of dexamethasone (S+D group), (six animal in each group). Total and differential white blood cell (WBC) counts in blood were evaluated. Oklahoma is one of three states to offer a free prekindergarten (pre-K) program to all students in participating school districts on a voluntary basis. Fortuitous circumstances in Tulsa, Oklahoma, the state's largest school district, permitted an unusually rigorous evaluation of the pre-K program in Tulsa. Because 4-year-olds beginning pre-K and 5-year-olds beginning kindergarten were administered the same test in September 2001 and because strict eligibility cutoffs were applied (based on date of birth), it was possible to control for selection effects, in addition to gender, race/ethnicity, school lunch eligibility, and precise date of birth. The evaluation showed strong positive effects of the pre-K program on children's language and cognitive test scores, but not on scores for social-emotional or motor skills. Hispanic children benefited most from the program, and Black children also showed sharp gains, especially when they attended full-day programs. In contrast, only those white children enrolled in a half-day pre-K program showed significant gains and only in language skills. A similar pattern of results characterized children who qualified for free lunch, for whom significant benefits derived from full- and part-day programs; children who qualified for a reduced price lunch, for whom only full-day programs produced gains; and children who did not qualify for lunch benefits, where no net gain was apparent. The findings were examined in light of contemporary controversies regarding targeted or universal pre-K programs; full- or part-day programming; public school or multiple delivery sites; strategies for ensuring high quality, effective programs; and the need for rigorous program evaluations. (saffron), which belongs to the Iridaceae family, is widely cultivated in Iran. Cardiovascular effects of saffron has been established in some studies but the effects of chronic administration of saffron (C. sativus) stigma aqueous extract on blood pressure has not been investigated. Objectives: In this study the effects of saffron (C. sativus) stigma aqueous extract on blood pressure of normotensive and desoxycorticosterone acetate (DOCA)-salt induced hypertensive rats, in chronic exposure was evaluated. Materials and Methods: Five weeks administration of three doses saffron aqueous extract (10, 20 and 40 mg/Kg/day) and spironolactone (50 mg/Kg/day) in different groups of normotensive and hypertensive rats (at the end of 4 weeks treatment by DOCA-salt) was carried out and their effects on mean systolic blood pressure (MSBP) and heart rate (HR) were evaluated using tail cuff method. The duration of the effect of saffron on systolic blood pressure (SBP), was also evaluated. Results: Our results indicated that chronic administration of saffron aqueous extract could reduce the MSBP in DOCA salt treated rats in a dose dependent manner. This compound did not decrease the MSBP in normotensive rats. The data also showed that antihypertensive effects of saffron did not persist. Conclusions: It is concluded that saffron aqueous extract possesses antihypertensive and normalizing effect on BP in chronic administration.

Introduction Saffron ( Crocus sativus L.) is a perennial plant of the Iridaceae family and is cultivated in Iran, Morocco, India, Greece, Italy, and Spain. Iran is considered the main producer of saffron, with the most optimum climate for saffron. Today, the dramatic increase in food production in the world requires high amounts of chemical fertilizers. However, some of the adverse effects of chemical fertilizer overuse in conventional farming practices have been well reported. In this regard, manure can be a good alternative, which can guarantee both agricultural production and nature protection. Potassium is one of the most important nutrients for plant growth after nitrogen. In spice plants, potassium indirectly improves nitrogen utilization and protein formation, flower size, flower yield, oil content, color, etc. in spice plants. This study investigated the effect of different animal manures in combination with different levels of potassium sulfate on saffron flower criteria. Materials and Methods The factorial experiment was carried out based on a randomized complete block design with three replications at the agricultural lands of Jovein County, during the three cropping years of 2015-16, 2016-17, and 2017-18. The first factor includes three types of organic manure: laying poultry manure (5 and 10 ton.ha -1 ), broiler poultry manure (5 and 10 t.ha -1 ) and cow manure (20 and 40 ton.ha -1 ) and control (without the use of organic manure) and the second factor includes different amounts of potassium sulfate fertilizer (0, 100 and 150 kg.ha -1 ). The basis for determining the consumption of each organic fertilizer was applied based on the recommended nitrogen and soil test. Saffron was cultivated in September 2015 in the length of 3 m and width of 1 m plots. Flower yield was determined randomly from 40 × 50 cm -2 quadrates. Data analysis and figure drawing were performed using SAS 9.1 and MS-Excel software. Results and Discussion The results of analysis of variance in two years showed that the effect of animal manure and potassium sulfate and their interaction were significant on the number of flowers, total flower weight, stigma dry weight, and style dry weight. Also, potassium sulfate and manure's main effects and interaction were significant on the dry weight of flowers without stigma and style in the second year. However, the main effects of potassium sulfate and manure significantly affected on the dry weight of flowers without stigma and style, but their interaction was not significant in the third year. The highest number of flowers was obtained in the treatment of 150 kg.ha -1 potassium sulfate and 40 ton.ha -1 cow manure in the second year, and it was obtained in the treatment of 150 kg.ha -1 potassium sulfate ten ton.ha -1 laying poultry manure in the third year. The highest total flower weight belonged to the treatment of 150 kg.ha -1 potassium sulfate and 10 ton.ha -1 was laying poultry manure in second and three years. The highest stigmas dry weight was observed in 100 kg.ha -1 potassium sulfate and 40 ton.ha -1 cow manure treatment in the second year, and the highest amount of this trait was related to the treatment of 100 kg.ha -1 potassium sulfate and ten ton.ha -1 broiler poultry manure in the third year. The results of this experiment indicate that the application of animal manures and potassium sulfate combination has positive effects on improving the yield of saffron flowers and eco-friendliness. It seems that the combination of animal manures and chemical fertilizers can reduce the use of chemical fertilizers. Conclusion The results of this study showed that the best treatment for most traits was 150 kg.ha -1 potassium sulfate and 40 ton.ha -1 cow manure interaction treatment in the second and third years, and then the 150 kg.ha -1 potassium sulfate and ten ton.ha -1 laying poultry manure treatment could be mentioned as the superior treatment. In general, according to the findings of this study, it seems that the simultaneous application of optimal amounts of ecological inputs and chemical fertilizers of potassium sulfate can reduce the destructive effects caused by excessive use of chemical fertilizers on this valuable medicinal plant while improving the properties of saffron flowers.
