This study aimed at developing a process for using local Ghanaian rice varieties to produce Aboloo; a traditional sweetened snack normally prepared from maize. Ten traditional producers of aboloo were interviewed to determine the critical unit operations and factors used in the preparation of aboloo to aid in process optimization. Consumer preference attributes for both baked and steamed aboloo were determined by interviewing 100 consumers using a semi-structured questionnaire. The magnitude of the preference for each attribute was determined using a 5 point scale (maximum = 5, minimum = 1). Critical unit operations identified that affect the quality included dehulling, soaking and fermentation. These unit operations were perceived by processors to affect the colour and texture of the final product. Four locally produced rice varieties, Beauty, Jasmine 85, Local Perfumed and Togo Marshall, were used for the process development of rice aboloo. For each rice variety, ten composite flour formulations involving rice, maize and wheat flour were made using the extreme vertices design. Sensory evaluation was carried by a trained panel to evaluate rice aboloo samples made from the four rice varieties. Rice aboloo made from Jasmine 85 with composite flour of 55% Jasmine85 rice: 40% maize: 5% wheat flour was found suitable for producing rice aboloo because the subsequent consumer acceptability test carried out on the developed steamed rice aboloo products was found to have the best desirable textural and sensory attributes.

Abstract The bacterial communities in floodwater, from a rice-planted and an unplanted field were characterized at the beginning (flooding stage) and at the end (harvest stage) of the rice cropping cycle. Most probable number estimations and plate counts of aerobic and anaerobic heterotrophic bacteria and of several metabolic bacterial groups (methanogens, sulfate-reducers, anaerobic sulfur and nonsulfur phototrophs, denitrifiers and ammonifiers) were similar in rice and unplanted floodwater at both sampling times. The analysis of denitrifiers and methanogens by fluorescent in situ hybridization revealed a shift in the phylogenetic affiliation only of the former group in the rice-planted floodwater. Terminal restriction fragment length polymorphism of 16S rRNA gene amplicons indicated that the bacterial communities of the rice-planted and unplanted soils were consistently diverse and strongly influenced by the season.

Abstract Background: Southern rice black-streaked dwarf virus (SRBSDV) is a recently discovered member of the genus Fijivirus and it is transmitted by the rice whitebacked planthopper (WBPH), Sogatella furcifera (Horváth). It was found that SRBSDV infected vectors might contribute negatively to the WBPH population, although the longer nymphal period might benefit viral acquisition, transmission and increase infection rate. The interaction between SRBSDV and its vector need to be further explored to gain better understanding of the dispersal of WBPH and the spread of virus disease, in particular the feeding and reproduction behavior of viruliferous WBPH. Methods: Newly hatched nymphs of WBPH were fed on healthy rice plant after feeding on SRBSDV-infected rice plants for 2 h, and newly emerged adults were numbered and tested. Feeding behaviors of WBPH adults were monitored electronically within a Faraday cage using a Giga-4 DC EPG amplifier. The newly emerged adults were paired, and the fecundity and egg hatchability were investigated. WBPH was molecularly identified for SRBSDV when they dead. According to the identification results, data on viruliferous and non-viruliferous WBPH were collected and analyzed. Results: Feeding behavior of viruliferous WBPH was different from those of non-viruliferous WBPH. Frequency of phloem sap ingestion of viruliferous WBPH increased significantly, however the total feeding duration did not increase markedly. When both WBPH parents were infected with SRBSDV, their fecundity and hatchability of the eggs produced were significant lower than those of normal WBPH parents. However, if only one of the parents was viruliferous, fecundity and egg hatchability were only slightly affected. Conclusions: Viruliferous WBPH fed on the phloem more frequently than non-viruliferous WBPH and can thus contribute to virus transmission. When both vector parents are viruliferous fecundity and hatchability of the eggs were significantly reduced. However when only one of the parents WBPH was viruliferous, there were no significant effects.

This article is from PLoS Genetics , volume 10 . Abstract Meiotic recombination normally takes place between allelic sequences on homologs. This process can also occur between non-allelic homologous sequences. Such ectopic interaction events can lead to chromosome rearrangements and are normally avoided. However, much remains unknown about how these ectopic interaction events are sensed and eliminated. In this study, using a screen in rice, we characterized a homolog of HUS1 and explored its function in meiotic recombination. In Oshus1 mutants, in conjunction with nearly normal homologous pairing and synapsis, vigorous, aberrant ectopic interactions occurred between nonhomologous chromosomes, leading to multivalent formation and subsequent chromosome fragmentation. These ectopic interactions relied on programed meiotic double strand breaks and were formed in a manner independent of the OsMER3-mediated interference-sensitive crossover pathway. Although early homologous recombination events occurred normally, the number of interference-sensitive crossovers was reduced in the absence of OsHUS1. Together, our results indicate that OsHUS1 might be involved in regulating ectopic interactions during meiosis, probably by forming the canonical RAD9-RAD1-HUS1 (9-1-1) complex.

This article is from Scientific Reports , volume 2 . Abstract The endoplasmic reticulum (ER) stress sensor, IRE1, contains a kinase domain and a ribonuclease domain. Ribonuclease mediates the unconventional splicing of mRNA encoding the transcription factor AtbZIP60 in Arabidopsis, or OsbZIP50 in rice, and thereby transduces signals from stressed ER. Here, we demonstrate the additional roles of plant IRE1 using genetically modified rice plants. Using a gene targeting system based on homologous recombination, genomic IRE1 was replaced with two types of missense alleles, leading to a defect in the kinase or ribonuclease activity of IRE1. Genetic analysis of these alleles demonstrated that the kinase activity of IRE1 plays a vital role independent of ribonuclease activity. Furthermore, the existence of ribonuclease substrates other than OsbZIP50 mRNA is demonstrated for the first time. This study provides new insights into higher plant signalling using a gene targeting approach.

This article is from BMC Genetics , volume 13 . Abstract Background: The Pi2/9 locus contains multiple nucleotide binding site–leucine-rich repeat (NBS-LRR) genes in the rice genome. Although three functional R-genes have been cloned from this locus, little is known about the origin and evolutionary history of these genes. Herein, an extensive genome-wide survey of Pi2/9 homologs in rice, sorghum, Brachypodium and Arabidopsis, was conducted to explore this theme. Results: In our study, 1, 1, 5 and 156 Pi2/9 homologs were detected in Arabidopsis, Brachypodium, sorghum and rice genomes, respectively. Two distinct evolutionary patterns of Pi2/9 homologs, Type I and Type II, were observed in rice lines. Type I Pi2/9 homologs showed evidence of rapid gene diversification, including substantial copy number variations, obscured orthologous relationships, high levels of nucleotide diversity or/and divergence, frequent sequence exchanges and strong positive selection, whereas Type II Pi2/9 homologs exhibited a fairly slow evolutionary rate. Interestingly, the three cloned R-genes from the Pi2/9 locus all belonged to the Type I genes. Conclusions: Our data show that the Pi2/9 locus had an ancient origin predating the common ancestor of gramineous species. The existence of two types of Pi2/9 homologs suggest that diversifying evolution should be an important strategy of rice to cope with different types of pathogens. The relationship of cloned Pi2/9 genes and Type I genes also suggests that rapid gene diversification might facilitate rice to adapt quickly to the changing spectrum of the fungal pathogen M. grisea. Based on these criteria, other potential candidate genes that might confer novel resistance specificities to rice blast could be predicted.

Introduction: Rice is the second most important edible grain after wheat in Iran. The most important factor for sustainable production in rice production lands, is water. Almost 75 percent of the world's rice is produced from paddy fields and rice is the largest consumer of water among all crops. Its growth is significantly affected by climate change and water scarcity. This research was carried out to compare the direct cultivation and transplanting of rice under different irrigation methods from the point of view of water productivity. Material and Methods: The current work was designed as split plot based on randomized complete block design with 6 treatments and three replications in 18 plots with 6 m wide and 12 m length (72 m 2 ) in area of approximately 1500 m 2 at the Agricultural Research Station of Golestan province in spring and the summer of 2018. Treatments including three levels of basin irrigation, sprinkler and drip (tape) irrigation were considered as main factors and two methods of direct cultivation and transplanting were considered as sub-factors. Results and Discussion : The results of analysis of variance showed that the effect of irrigation and sowing method on the yield of rice were significant at 0.01 level probability. The highest amount of biological yield in transplanting was obtained by basin irrigation (8177 kg/ha) and in direct seeding in basin irrigation and taper irrigation (7375 and 6836 kg/ha, respectively). The highest 1000-grain weight in transplanting method was obtained in basin irrigation and direct seeding method in traditional irrigation and tape irrigation. The highest number of filled grains in the panicle was observed in the basin irrigation treatment in transplanting, with significant difference compared to the amount observed in direct seeding. Traditional irrigation between planting methods had not significant effect on the number of hollow grains; while in sprinkler and tape irrigation (with an average of 51 and 56 for sprinkler and tape irrigation, respectively), the number of hollow grains had a higher rate in direct seeding (with an average of 41 and 45 for sprinkler and tape irrigation). The results showed that basin irrigation with 8177 kg/ha grain yield in transplanting method and basin and tape irrigations with 7375 and 6836 kg/ha in direct seeding method had the highest grain yield. Sprinkler irrigation had the lowest paddy yields in transplanting (4188 kg/ha) and direct seeding (5712 kg/ha). Tape irrigation with 7390 and 6840 m 3 of consumed water, resulted in lower water consumption compared to traditional irrigation (10700 and 1693 m 3 ), respectively. The highest amount of water use efficiency was obtained in direct and tape irrigation (0.99 kg/m 3 ) and in traditional and tape irrigation (0.76 and 0.66 kg/m 3 , respectively). Conclusion: Adjusting water consumption both through lower water consumption and reduced water wastage can lead higher water productivity of rice production systems creating sustainable rice production systems. In transplanting method, tape irrigation and traditional irrigation had higher water use efficiency than sprinkler irrigation, although a significant water use reduction in tape irrigation rather than basin irrigation should be analyzed economically at real water prices. Overall, in this study, the tape irrigation method, by reducing water consumption, was able to increase water productivity and maintain the yield of rice in direct cultivation. Sprinkler irrigation had the lowest yield of rice in transplanting and direct cultivation. Problems such as lack of uniformity of spraying and waste water through the wind as well as poor quality of sprinklers are the main reasons for the decrease of the efficiency of sprinkler irrigation.
