This systematic review and meta-analysis aims to evaluate the effect of vitamin D on left ventricular (LV) geometry among individuals with primary hypertension. Specifically, it investigates two related research questions: (1) whether vitamin D supplementation alters LV geometric parameters in clinical trials involving hypertensive patients, and (2) whether serum vitamin D deficiency is associated with abnormal LV geometry in observational studies. LV geometric outcomes of interest include LV mass, volume, wall thickness, ejection fraction, and derived indices such as LV mass index. By pooling data from both interventional and observational studies, this project seeks to determine the strength and direction of associations between vitamin D levels and structural cardiac changes in hypertension. This review adheres to the PRISMA reporting guidelines. The literature search was conducted across Scopus, PubMed, EuropePMC, Web of Science, and Scilit up to June 7, 2024, without language or publication-type restrictions. Eligible studies included randomized controlled trials, non-randomized trials, and cross-sectional studies, provided they reported quantitative data on LV geometry in adult patients with primary hypertension. Studies involving participants with secondary hypertension, chronic kidney disease, or other conditions affecting vitamin D metabolism were excluded. The expected outcomes of this review are to (1) clarify the extent to which vitamin D supplementation may reverse or prevent adverse cardiac remodeling in hypertension, and (2) identify whether vitamin D deficiency contributes to subclinical cardiac structural changes. These findings may support future recommendations for vitamin D monitoring or supplementation in the management of hypertensive heart disease.

Abstract The initial observations linking vitamin D to type 2 diabetes in humans came from studies showing that both healthy and diabetic subjects had a seasonal variation of glycemic control. Currently, there is evidence supporting that vitamin D status is important to regulate some pathways related to type 2 diabetes development. Since the activation of inflammatory pathways interferes with normal metabolism and disrupts proper insulin signaling, it is hypothesized that vitamin D could influence glucose homeostasis by modulating inflammatory response. Human studies investigating the impact of vitamin D supplementation on inflammatory biomarkers of subjects with or at high risk of developing type 2 diabetes are scarce and have generated conflicting results. Based on available clinical and epidemiological data, the positive effects of vitamin D seem to be primarily related to its action on insulin secretion and sensitivity and secondary to its action on inflammation. Future studies specifically designed to investigate the role of vitamin D on type 2 diabetes using inflammation as the main outcome are urgently needed in order to provide a more robust link between vitamin D, inflammation and type 2 diabetes.

Calcitriol plays a critical role in maintaining mineral homeostasis but also exhibits antiproliferative activity in many cancers. We have shown that the antiproliferative actions of calcitriol in the LNCaP human prostate cancer (PCa) cell is mediated in large part by induction of insulin-like growth factor binding protein-3 (IGFBP-3). The purpose of this study was to determine the molecular mechanism involved in calcitriol regulation of IGFBP-3 and to identify the putative vitamin D response element (VDRE) in the IOFBP-3 promoter. We cloned 6 kb of the IGFBP-3 promoter and demonstrated its responsiveness to calcitriol in transactivation assays. Computer analysis identified a putative VDRE between -32961/-3282 that is similar to other known VDREs. In gel shift assays the vitamin D receptor (VDR) showed strong calcitriol-dependent binding to this putative VDRE. ChIP assay demonstrated that calcitriol recruited the VDR/RXR heterodimer to the VDRE site. In transactivation as says the VDRE promoter was induced 2-fold by calcitriol. Mutations created in the VDRE resulted in a loss of IGFBP-3 induction confirming the critical VDRE sequence. In conclusion, we have identified a functional VDRE in the distal region of the human IGFBP-3 promoter that directly mediates the action of calcitriol.

The most significant achievement of this period is the development of a synthetic scheme to produce substantial quantity of our target compound i.e. 1,25- dihydroxyvitamin D3-3-bromoacetate. This is extremely important for the current project and for the future development of this compound for prostate cancer. We have also screened this compound against prostate and kidney cancer cells for its antiproloferative activity. In addition we have developed a mouse xenograft model to test the efficacy of this compound in reducing androgen-sensitive and androgen-insensitive prostate tumors in future studies.

Vitamin D and its metabolites have clinical significance because they play a critical function in calcium homeostasis and bone metabolism. Although not all of the pathologic mechanisms have been adequately described, vitamin D insufficiency and deficiency, as measured by low levels of 25-OH vitamin D, are associated with a variety of clinical conditions including osteoporosis, falls and fractures in the elderly, decreased immune function, bone pain, and possibly colon cancer and cardiovascular health.2 Apart from inadequate dietary intake, patients may present with low levels of vitamin D if they receive inadequate sunlight. The astronaut population is potentially vulnerable to low levels of vitamin D for several reasons. Firstly, they may train for long periods in Star City, Russia, which by virtue of its northern latitude receives less sunlight in winter months. Secondly, astronauts are deprived of sunlight while aboard the International Space Station (ISS). In addition, ISS crew members are exposed to microgravity for prolonged durations and are likely to develop low bone mineral density despite the use of countermeasures. Therefore, closely monitoring and maintaining adequate vitamin D levels is important for the astronaut corps.

ABSTRACT Aims: The Covid-19 pandemic started in Wuhan City, Hubei Province, China in late December 2019. In our study, we aimed to evaluate the effectiveness of vitamin D levels on the clinic of the disease, laboratory findings, severity of the disease and, length of hospital stay by grouping pregnant patients with a diagnosis of Covid-19 according to their vitamin D levels. Methods: A total of 125 patients were included. According to the vitamin D levels of the patients at the time of hospitalization, two groups were determined as below and above 20ng/ml, which is the limit of vitamin D deficiency. The patients in these two groups were compared in terms of demographic features, clinical findings, laboratory findings, imaging findings, hospitalization times and need for intensive care. Results: When evaluated according to serum 25 hydroxyvitamin D level, patients’ fever, pulse, oxygen saturation, the severity of lung involvement in computed tomography, hospitalization there was no statistically significant difference between the groups in terms of duration and need for intensive care. When the laboratory parameters of the patients at hospitalization were compared according to serum 25 hydroxyvitamin D levels, there was no statistically significant difference between the two groups in terms of any laboratory parameter. As a result of the analysis, only lymphocyte count was determined as the independent variable affecting the severity of lung involvement in thorax CT (Computed tomography). As the lymphocyte count decreased, the severity of involvement in thorax CT increased. Conclusion: Our study showed that vitamin D level did not have a significant relationship with any of the parameters related to Covid-19 such as clinical and laboratory findings, severity of the disease and duration of hospitalisation. It is also supported by our study that the decrease in lymphocyte counts is associated with severe Covid-19 disease.

The influence of the submarine environment on absorption of ingested vitamin D has not been previously investigated. This study examines the vitamin D absorption patterns of 5 subjects at the beginning, middle, and end of a 69 day patrol on a normal diet and with no exposure to sunlight. It was found that the magnitude and pattern of absorption does not change appreciably. It is concluded that the absorption of vitamin D is not affected by the submarine environment and that any drop in 25(OH) vitamin D levels seen during patrol is caused by lack of sunlight, ultraviolet light in particular. Implications in terms of sunlight deprivation and calcium metabolism are discussed in a historical review of calcium and vitamin D investigations on submarines.

Abstract As skin cancer is one of the most costly health issues in many countries, particularly in Australia, the possibility that vitamin D compounds might contribute to prevention of this disease is becoming increasingly more attractive to researchers and health communities. In this article, important epidemiologic, mechanistic and experimental data supporting the chemopreventive potential of several vitamin D-related compounds are explored. Evidence of photoprotection by the active hormone, 1α,25dihydroxyvitamin D3, as well as a derivative of an over-irradiation product, lumisterol, a fluorinated analog and bufalin, a potential vitamin D-like compound, are provided. The aim of this article is to understand how vitamin D compounds contribute to UV adaptation and potentially, skin cancer prevention.

Background: Socioeconomics only account for 18% of all healthcare disparities. Healthcare disparities in the intensive care unit (ICU) have been well documented and persist in spite of previous government and medical interventions. Vitamin D deficiency is the most common nutritional deficiency in the United States and the world. This deficiency has been largely overlooked in the debate on healthcare disparities.

Originally released on May 16, 2016 Amy speaks with Don Bennett who is an independent researcher, Disease Avoidance Specialist, director of  Health 101 , author, and coach. At a young age, Don discovered inconsistencies within the health field. Being a seeker of truth, he set out to learn the realities of health for himself. Discovering that traditional curriculums were filled with biases, hidden agendas, misinformation, and missing information, Don remained self-taught, examining info as a researcher and not studying it as a student, thereby avoiding the hurdles set up by academic tradition. The experiments in Round 2 were designed to extend the results of our efforts in Round 1 which led us to hypothesize that the seco-steroid, 1,25-dihydroxyvitamin D3[1,25(OH)2D3], acts in synergy with parathyroid hormone (PTH) to regulate bone calcium homeostasis. Our work centered on one particular target of 1,25(OH)2D3 action, the voltage-sensitive calcium channels (VSCC's), which are activated acutely by this steroid within milliseconds of exposure . A second area of research focused on the effects of mechanical strain on VSCC expression in bone. These experiments were performed in collaboration with Dr. Steven Goldstein (Univ. Michigan), who generously provided RNA extracted from dog bones that had been exposed to mechanical strain in vivo. Our results suggest that mechanical loading elevated VSCC expression in the long bones from 3 of the 6 animals tested. A second line of experimentation, carried out in collaboration with Dr. Randall Duncan, a NASA-funded investigator in Indianapolis, centered on RT-PCR analysis of effects of mechanical strain on Ca2(+) channel expression in cultured bone cells. Compared to unstrained controls, the expression of vitamin-D-sensidve Ca2(+) channels is elevated 3- to 5-fold over a 24 hr period.

This article is from PLoS ONE , volume 9 . Abstract Environmental factors such as diet, intake of vitamin D supplements and exposure to sunlight are known to influence serum vitamin D concentrations. Genetic epidemiology of vitamin D is in its infancy and a better understanding on how genetic variation influences vitamin D concentration is needed. We aimed to analyse previously reported vitamin D-related polymorphisms in relation to serum 25(OH)D concentrations in 201 healthy Danish families with dependent children in late summer in Denmark. Serum 25(OH)D concentrations and a total of 25 SNPs in GC, VDR, CYP2R1, CYP24A1, CYP27B1, C10or88 and DHCR7/NADSYN1 genes were analysed in 758 participants. Genotype distributions were in Hardy–Weinberg equilibrium for the adult population for all the studied polymorphisms. Four SNPs in CYP2R1 (rs1562902, rs7116978, rs10741657 and rs10766197) and six SNPs in GC (rs4588, rs842999, rs2282679, rs12512631, rs16846876 and rs17467825) were statistically significantly associated with serum 25(OH)D concentrations in children, adults and all combined. Several of the SNPs were in strong linkage disequilibrium, and the associations were driven by CYP2R1-rs10741657 and rs10766197, and by GC-rs4588 and rs842999. Genetic risk score analysis showed that carriers with no risk alleles of CYP2R1-rs10741657 and rs10766197, and/or GC rs4588 and rs842999 had significantly higher serum 25(OH)D concentrations compared to carriers of all risk alleles. To conclude, our results provide supporting evidence that common polymorphisms in GC and CYP2R1 are associated with serum 25(OH)D concentrations in the Caucasian population and that certain haplotypes may predispose to lower 25(OH)D concentrations in late summer in Denmark.

Abstract This study aimed to investigate the association between serum levels of 25-hydroxyvitamin D [25(OH)D] and metabolic syndrome along with its associated risk factors in Korean postmenopausal women. This study was performed using data from the KNHANES 2008–2010 study and included 4,364 postmenopausal Korean women. Clinical and other objective characteristics, seasonality, and presence of metabolic syndrome with its five components were evaluated and correlated with the serum levels of 25(OH)D. Although no statistically significant associations were observed between the levels of serum 25(OH)D and the prevalence of metabolic syndrome, the adjusted OR for elevated blood pressure, elevated triglycerides (TGs), and reduced high-density lipoprotein cholesterol (HDL-C) showed tendency to decrease sequentially as tertiles of serum 25(OH)D levels increased (p for trends  = 0.066, 0.043, and 0.010, respectively). Women in the highest tertile of serum 25(OH)D showed a significant decrease in the prevalence of elevated blood pressure, elevated TGs, and reduced HDL-C as compared with those in the lowest tertile of serum 25(OH)D (p = 0.020, 0.014, and 0.002, respectively). Based on these results, we consider that adequate serum levels of 25(OH)D in Korean postmenopausal women may not entirely indicate a lower risk of developing metabolic syndrome. However, adequate serum levels of 25(OH)D are significantly associated with a decrease in elevated blood pressure, elevated TGs, and reduced HDL-C levels in postmenopausal women.

Abstract Central giant cell granulomas (CGCGs) are uncommon but the most aggressive benign intraosseous tumors of jaws, with an unpredictable outcome. They account for less than 7% of all benign jaw lesions, with a female to male ratio of about 2:1. The classical “brown tumor” is commonly seen in the long bones, pelvis, and ribs. Facial bone involvement is rare and usually appears as solitary or multilocular soap bubble like radiolucencies. CGCGs are traditionally treated by both surgical and intralesional injection, with a variable recurrence rate. Here, we report a 12-year-old female patient with mandibular brown tumor as a first sign of secondary hyperthyroidism induced due to vitamin D deficiency and hypocalcemia.

Methods:: This cross-sectional study was carried out on 254 (128 male and 126 female) university students. Demographic questionnaires and a questionnaire on exposure to sun light and sun protection were completed by the participants. Serum 25OH-vitamin D was measured using a radioimmunoassay kit. Data analysis was done using Statistical Package for Social Sciences (SPSS) software # 16. A P value less than 0.05 was considered as significant. Results:: Mean ± standard deviation (SD) of serum 25OH-vitamin D was 49.29 ± 12.87 (nmol/l) and 27.46 ± 10.37 (nmol/l) among male and female students, respectively. 51.2% of female students were vitamin D insufficient and 44% of them had vitamin D deficiency. Prevalence of vitamin D insufficiency and marginal status among male students were 49.5 and 48%, respectively. Serum vitamin D of female students was significantly less than the males (P < 0.001). Serum vitamin D was negatively correlated to sun protection score (P < 0.001, r = 0.50), but there was no correlation between serum vitamin D and sun exposure. Conclusions:: Vitamin D deficiency especially among female students is alarmingly prevalent. Increasing use of sunscreen lotion and clothing style could be the main factors inhibiting endogenous vitamin D synthesis which results in its deficiency.

Endothelial colony forming cells (ECFC) are endothelial progenitor cells that participate in vasculogenesis and endothelial repair. Objective: We hypothesized that the number and functional properties of fetal cord blood-derived ECFCs are reduced in preeclampsia compared to uncomplicated pregnancy (controls), and asked if adverse effects of preeclampsia on ECFC function are reversed by 1,25 (OH)2 vitamin D3. Design, Setting, Patients: This was a nested, case-control study. Forty women with uncomplicated pregnancy and 33 women with PE were recruited at Magee-Womens Hospital (USA) or at Hannover Medical School (Germany). Main Outcome Measures: Time to ECFC colony appearance in culture, and number of colonies formed, were determined. Functional abilities of ECFCs were assessed in vitro by tubule formation in Matrigel assay, migration, and proliferation. ECFC function was tested in the presence or absence of 1,25 (OH)2 vitamin D3, and after vitamin D receptor (VDR) or VEGF signaling blockade. Abstract Changes in vitamin D serum levels have been associated with inflammatory diseases, such as inflammatory bowel disease (IBD), rheumatoid arthritis, systemic lupus erythematosus, multiple sclerosis (MS), atherosclerosis, or asthma. Genome- and transcriptome-wide studies indicate that vitamin D signaling modulates many inflammatory responses on several levels. This includes (i) the regulation of the expression of genes which generate pro-inflammatory mediators, such as cyclooxygenases or 5-lipoxygenase, (ii) the interference with transcription factors, such as NF-κB, which regulate the expression of inflammatory genes and (iii) the activation of signaling cascades, such as MAP kinases which mediate inflammatory responses. Vitamin D targets various tissues and cell types, a number of which belong to the immune system, such as monocytes/macrophages, dendritic cells (DCs) as well as B- and T cells, leading to individual responses of each cell type. One hallmark of these specific vitamin D effects is the cell-type specific regulation of genes involved in the regulation of inflammatory processes and the interplay between vitamin D signaling and other signaling cascades involved in inflammation. An important task in the near future will be the elucidation of the regulatory mechanisms that are involved in the regulation of inflammatory responses by vitamin D on the molecular level by the use of techniques such as chromatin immunoprecipitation (ChIP), ChIP-seq, and FAIRE-seq.
