This article is from Mobile Genetic Elements , volume 4 . Abstract Transposons are mobile genetic elements that can be harmful for the host when mobilized. However, they are also genomic reservoirs for novel genes that can be evolutionarily beneficial. There are many examples of domesticated transposases, which play important roles in the hosts. In most cases domesticated transposases have lost their endonuclease activities and the hosts utilize their DNA-binding properties. However, some other domesticated transposases perform endonuclease activities for host biological processes. Because such a catalytically active transposase is potentially harmful for the integrity of the host genome, its activity should be tightly regulated. The catalytically active domesticated piggyBac transposase Tpb2p catalyzes programmed DNA elimination in the ciliate Tetrahymena. Here, we discuss the regulatory mechanism that prevents unintended DNA cleavage by Tpb2p and compare it to another well-studied catalytically active domesticated transposase, the RAG recombinase in V(D)J recombination. The regulatory mechanisms involve the temporarily regulated expression of the transposases, the target sequence preference of the endonuclease, and the recruitment of the transposases to locally restricted chromatin environments.

By Andre Sobocinski, historian, U.S. Navy Bureau of Medicine and Surgery Wendell Phillips’s ambitious plan of exploration from “Cairo to Capetown” offered Navy Medicine a veritable “lost world” of opportunity to investigate indigenous diseases and collect teaching specimens. Five months later, the newly formed “Navy unit” headed by tropical medical specialist Cmdr. (later Capt.) Julius Amberson, MC, USN,4 sailed from New York to join the UC Berkeley contingent in Cairo. Amberson’s team was comprised of a host of eminent military and civilian parasitologists, zoologists, as well as a preventive medical technician and a medical photographer. The unit was furnished with its own trucks, jeeps, radio and photographic equipment, guns and  ammunition, an X-ray machine, and the latest in laboratory equipment.

This personal anthology is my choice of speeches from Shakespeare that I enjoy reading (that I would like to have had by heart years ago!) and that seem to me to illustrate his unsurpassed use of language. He was a man who seemed to know everything about human nature and as Orson Welles said ‘he speaks to everyone and we all claim him’. I know that it has been said that ‘it is impossible to be a great Shakespearian actor without an idiosyncratic and extraordinary voice’ and this may be so, but that does not preclude ordinary mortals from reading, hearing and enjoying Shakespeare. LibriVox volunteers bring you seventeen recordings of Sonnet 116 by William Shakespeare. This was the weekly poetry project for the week of October 15th, 2006.

<i>King Henry IV, Part 1</i> is the second of Shakespeare’s eight Wars of the Roses history plays, with events following those of <i>King Richard II</i>. As the play opens, King Henry IV (formerly Henry Bolingbroke) and Henry Percy (Hotspur) argue over the disposition of prisoners from the Battle of Holmedon. The King’s attitude toward Mortimer and the Percy family prompts them to plot rebellion. In the meantime, his son Prince Hal is living the low life in the company of Sir John Falstaff. As the time of battle nears, Prince Hal joins his father and is given a high command. The play’s climax is the Battle of Shrewsbury, in which Prince Hal and Hotspur meet and fight, with Prince Hal and the forces of the King prevailing. Written around the middle of his career, <i>Much Ado About Nothing</i> is one of Shakespeare's great festive comedies. The men are back from the war, and everyone is ready for romance. The dashing young Claudio falls for Hero, the daughter of Leonato, governor of Messina, and his friend Don Pedro helps him secure her affection. These youthful lovers are contrasted with the more experienced (and more cynical) Benedick and Beatrice, who have to be tricked into falling in love. LibriVox readers present the seventh collection of monologues from Shakespeare’s plays. Containing 20 parts. <br /> William Shakespeare (April 26, 1564 – April 23, 1616) remains widely to be considered the single greatest playwright of all time. He wrote in such a variety of genres - tragedy, comedy, romance, &c - that there is always at least one monologue in each of his plays. Some of these teach a lesson, some simply characterize Shakespeare at his best, some are funny, some sad, but all are very moving. Each monologue will touch everybody differently. Magic, fairies, young lovers chasing each other through a forest, a man with a donkey's head, and impish Puck wreaking havoc right and left. What's going on here? It's <em>A Midsummer Night's Dream</em>, Shakespeare at his most fanciful. The play opens with Theseus, Duke of Athens, preparing for his wedding. Egeus complains to Theseus that his daughter Hermia refuses to marry Demetrius. When Hermia is given the choice between marriage to Demetrius or life as a nun, she and her true love Lysander flee into the forest. Demetrius follows them; and Helena, who loves Demetrius, follows <em>him</em>. Also in the forest are Oberon and Titania, king and queen of the fairies, at odds with one another. At Oberon's behest, Puck causes Demetrius to fall in love with Helena -- oops, he missed, that was Lysander instead. Mayhem ensues. In the meantime, a group of bumbling craftsmen rehearses a play. Puck gives one of them, Bottom, the head of an ass and makes Titania fall in love with him. Further hilarity results as Bottom sees nothing at all odd about this. Eventually everything is straightened out, Bottom and the rest "perform" their play, there is a triple wedding, and Puck assures us the whole thing has been a dream.
