This article is from BMC Developmental Biology , volume 14 . Abstract Background: The Drosophila heart (dorsal vessel) is a relatively simple tubular organ that serves as a model for several aspects of cardiogenesis. Cardiac morphogenesis, proper heart function and stability require structural components whose identity and ways of assembly are only partially understood. Structural components are also needed to connect the myocardial tube with neighboring cells such as pericardial cells and specialized muscle fibers, the so-called alary muscles. Results: Using an EMS mutagenesis screen for cardiac and muscular abnormalities in Drosophila embryos we obtained multiple mutants for two genetically interacting complementation groups that showed similar alary muscle and pericardial cell detachment phenotypes. The molecular lesions underlying these defects were identified as domain-specific point mutations in LamininB1 and Cg25C, encoding the extracellular matrix (ECM) components laminin β and collagen IV α1, respectively. Of particular interest within the LamininB1 group are certain hypomorphic mutants that feature prominent defects in cardiac morphogenesis and cardiac ECM layer formation, but in contrast to amorphic mutants, only mild defects in other tissues. All of these alleles carry clustered missense mutations in the laminin LN domain. The identified Cg25C mutants display weaker and largely temperature-sensitive phenotypes that result from glycine substitutions in different Gly-X-Y repeats of the triple helix-forming domain. While initial basement membrane assembly is not abolished in Cg25C mutants, incorporation of perlecan is impaired and intracellular accumulation of perlecan as well as the collagen IV α2 chain is detected during late embryogenesis. Conclusions: Assembly of the cardiac ECM depends primarily on laminin, whereas collagen IV is needed for stabilization. Our data underscore the importance of a correctly assembled ECM particularly for the development of cardiac tissues and their lateral connections. The mutational analysis suggests that the β6/β3/β8 interface of the laminin β LN domain is highly critical for formation of contiguous cardiac ECM layers. Certain mutations in the collagen IV triple helix-forming domain may exert a semi-dominant effect leading to an overall weakening of ECM structures as well as intracellular accumulation of collagen and other molecules, thus paralleling observations made in other organisms and in connection with collagen-related diseases.

3Application Of The Effective Fisher Matrix To The Frequency Domain Inspiral Waveforms

The Fisher matrix (FM) has been generally used to predict the accuracy of the gravitational wave parameter estimation. Although a limitation of the FM has been well known, it is still mainly used due to its very low computational cost compared to the Monte Carlo simulations. Recently, Rodriguez et al. [Phys. Rev. D 88, 084013 (2013)] performed Markov chain Monte Carlo (MCMC) simulations for nonspinning binary systems with total masses $M \leq 20 M_{\odot}$, they found systematic differences between the predictions from FM and MCMC for $M>10 M_{\odot}$. On the other hand, an effective Fisher matrix (eFM) was recently introduced by Cho et al. [Phys. Rev. D 87, 24004 (2013)]. The eFM is a semi-analytic approach to the standard FM, in which the partial derivative is taken by a quadratic fitting function to the local overlap surface. In this work, we apply the eFM method to several nonspinning binary systems and find that the error bounds in eFM are qualitatively in good agreement with the MCMC results of Rodriguez et al. in all mass regions. In particular, we provide concrete examples showing an importance of taking into account the template-dependent frequency cutoff of the inspiral waveforms.

Abstract Background: Transglutaminase-4 (TGase-4), also known as the Prostate Transglutaminase, is an enzyme found to be expressed predominately in the prostate gland. The protein has been recently reported to influence the migration and invasiveness of prostate cancer cells. The present study aimed to investigate the influence of TGase-4 on cell-matrix adhesion and search for the candidate active domain[s] within the protein. Methods: Human prostate cancer cell lines and prostate tissues were used. Plasmids that encoded different domains and full length of TGase-4 were constructed and used to generate sublines that expressed different domains. The impact of TGase-4 on in vitro cell-matrix adhesion, cell migration, growth and in vivo growth were investigated. Interactions between TGase-4 and focal adhesion complex proteins were investigated using immunoprecipitation, immunofluorescence and phosphospecific antibodies. Results: TGase-4 markedly increased cell-matrix adhesion and cellular migration, and resulted in a rapid growth of prostate tumours in vivo. This effect resided in the Core-domain of the TGase-4 protein. TGase-4 was found to co-precipitate and co-localise with focal adhesion kinase (FAK) and paxillin, in cells, human prostate tissues and tumour xenografts. FAK small inhibitor was able to block the action mediated by TGase-4 and TGase-4 core domain. Conclusion: TGase-4 is an important regulator of cell-matrix adhesion of prostate cancer cells. This effect is predominately mediated by its core domain and requires the participation of focal adhesion complex proteins.

7Nature Of Matrix Elements In The Quantum Chaotic Domain Of Interacting Particle Systems

There is a newly emerging understanding that in the chaotic domain of isolated finite interacting many particle systems smoothed densities define the statistical description of these systems and these densities follow from embedded (two-body) random matrix ensembles and their various deformations. These ensembles predict that the smoothed form of matrix elements of a transition operator between the chaotic eigenstates weighted by the densities at the two ends (i.e. the bivariate strength density) will be a bivariate Gaussian with the bivariate correlation coefficient arising out of the non-commutability of the hamiltonian and the transition operator involved. The ensemble theory extends to systems with a mean-field and a chaos generating two-body interaction (as in nuclei, atoms and diffusive quantum dots). These developments in many-body quantum chaos are described with special reference to one-body transition operators.

We report on our computation of the hadronic matrix elements of the four-quark operators needed for the study of $K^0-{\bar K^0}$ mixing beyond the Standard Model (SM). We consider nf=2+1 Domain-Wall fermions on Iwasaki gauge action with lightest unitary pion of 290 MeV and a single lattice spacing a=0.086 fm. The renormalization is performed non-perturbatively through the RI-MOM scheme and our results are converted perturbatively to MSbar. We have estimated the various systematic errors. Our results confirm a previous quenched study, where large ratios of non-SM to SM matrix elements were obtained.

10Oscillatory Terms In The Domain Wall Transfer Matrix

We study the transfer matrix for domain wall fermions to understand the origin and significance of oscillatory contributions to hadron correlation functions that arise for M >1. For a free particle in one space, one time, and one flavor dimension, the eigenmodes of the one-body operator appearing in the transfer matrix are calculated, and the role of the negative eigenmodes arising when M > 1 is studied. In the case of three space dimensions, oscillatory behavior for hadron correlation functions in QCD is shown to emerge for free fermions when M exceeds 1, and to increase with increasing M. Analogous behavior is observed for domain wall fermions on HYP smeared MILC lattices, and a procedure is demonstrated for subtracting oscillating terms from physical observables.
