A simple method is proposed for the determination of effective rain cell dimensions and orientation. Two rain cell models were considered: the circular cell and the elliptical cell. In both models it is assumed that the rain rate is constant throughout the cell, that all cell locations are equally likely, and that the cell dimensions depend upon the rain rate. It is then shown that the effective cell dimensions and orientation may be deduced from rain rate statistics accumulated at two or three closely spaced sites, depending upon the model chosen. The results of this study alone with previous estimates of rain cell dimension indicate that the rain rate measurement points should be spaced on the order of one-half kilometer apart. The proposed method is simple and relatively inexpensive; thus, this approach is readily suited to the study of the dependence of rain cell size and orientation upon climatic region. The resulting rain cell characteristics are of direct value in the prediction of millimeter wavelength attenuation statistics on both single-terminal and diversity earth-space propagation paths as well as point-to-point terrestrial links.

The closed-cycle hydrogen-oxygen PEM regenerative fuel cell (RFC) at the NASA Glenn Research Center has successfully demonstrated closed cycle operation at rated power for multiple charge-discharge cycles. During charge cycle the RFC has absorbed input electrical power simulating a solar day cycle ranging from zero to 15 kWe peak, and delivered steady 5 kWe output power for periods exceeding 8 hr. Orderly transitions from charge to discharge mode, and return to charging after full discharge, have been accomplished without incident. Continuing test operations focus on: (1) Increasing the number of contiguous uninterrupted charge discharge cycles; (2) Increasing the performance envelope boundaries; (3) Operating the RFC as an energy storage device on a regular basis; (4) Gaining operational experience leading to development of fully automated operation; and (5) Developing instrumentation and in situ fluid sampling strategies to monitor health and anticipate breakdowns.

Our patch clamp studies indicate MscCa is expressed by the invasive prostate tumor cell PC-3. Anti-MscCa agents, Gd3+, GsmTx-4, and an anti-TRPC1 antibody block PC-3 cell migration. MscCa activity can be recorded over the surface of the PC-3 cell but is expressed at higher density on the rear compared with the front of the cell. This channel density gradient combined with a higher density of thapsigargin-sensitive Ca2+ stores in the rear of the cell enables the development of an intracellular Ca2+ gradient (low front high rear) in migrating PC- 3 cells that determines migration directionality. Gene silencing of TRPC1 and/or TRPC3, but not TRPC4 or TRPC6, blocks PC-3 cell migration. Permanently suppressing TRPC1 also reduces PC-3 cell proliferation and thereby blocks tumor invasion in vivo. The noninvasive human prostate tumor cell line LNCaP expresses MscCa but the channel undergoes rapid inactivation that prevents Ca2+ gradient development and directional cell migration. Our results indicate that specific forms of mechanical stimuli can switch the inactivating gating mode to the non-inactivating mode seen in PC-3 cells, and this switch is independent of the actin-cytoskeleton. These findings have specific implications regarding the possible role of the increases mechanical forces (e.g., solid stress and interstitial fluid compression) that develop within a growing prostate tumor in promoting its progression to malignancy.

SAMPLE CELL GERMANIUM OVERFLOW OF THE DIFFUSION PROCESS IN MOLTEN SEMICONDUCTOR FLIGHT EXPERIMENT DPIMS

This is a retrospective real-world data science study. By leveraging real-world data from China, including electronic medical records (EMR) and healthcare claims data, we systematically assess the ability of non-randomized RWE to replicate multiple RCTs, and gain a deeper understanding of the advantages and limitations of using non-randomized RWE in regulatory decision-making. All patients who visited the selected centers from January 1, 2019 to November 30, 2024 and met the inclusion and exclusion criteria were included. Demographic characteristics, clinical features, treatment patterns and prognosis of the patients were collected.

Results of the Independent Orbiter Assessment (IOA) of the Failure Modes and Effects Analysis (FMEA) and Critical Items List (CIL) are presented. The IOA effort first completed an analysis of the Electrical Power Generation/Fuel Cell Powerplant (EPG/FCP) hardware, generating draft failure modes and potential critical items. To preserve independence, this analysis was accomplished without reliance upon the results contained within the NASA FMEA/CIL documentation. The IOA results were then compared to the proposed Post 51-L NASA FMEA/CIL baseline. A resolution of each discrepancy from the comparison was provided through additional analysis as required. This report documents the results of that comparison for the Orbiter EPG/FCP hardware.

Work has continued on the Top Down approach to genome minimization. Because the smallest genome to date grows too slowly to be used in future experiments, we have conducted additional transposon studies and used the data to construct a new, fast growing parental strain. The effort to modularize the genome has been initiated. We have made a modular map of the genome by grouping genes according to function. In the future, we will modularize the genome as much as possible, although we expect that there will be some general genome layout requirements that will make full modularization impossible. Preliminary work aimed at genome complementation has been conducted. A plasmid system for quickly adding back deleted genes will allow us to quickly examine which gene(s) within a deleted cluster might be causing growth defects or cell death.

The method can be used in any test cell or an outdoor site and should be considered as a good alternative to test cell correlations. Estrogen affects both the cytoskeletal and chromatin structure of breast cancer cells. The treatment of breast cancer cells with 10 nM estradiol causes the estrogen receptor to recruit histone acetyltransferases to the promoter of the estrogen-responsive PS2 gene within 60 min, while exposure to estrogen for 72 h increases the levels of DNA-associated cytokeratins. To determine the effect of estrogen on regions downstream of estrogen- responsive promoters, quantitative PCR was performed on DNA isolated from chromatin immunoprecipitations using anti-acetylated 113 and H4 antibodies. Treatment with 10 nM estradiol induced a transient state of 113 and 114 acetylation along the PS2 promoter, exon 2, and exon 3, that was maximal after 60-120 min. After three hours, this hyperacetylation decreased to levels above those observed along the PS2 gene in untreated MCF-7 cells. To determine how estrogen influences DNA-associated cytokeratins, the levels of total, intermediate filament-assembled and DNA-associated cytokeratins were determined by Western Blotting and two dimension gel electrophoresis. The level of DNA-associated cytokeratins increased after 2 h of 10 nM estradiol treatment, while the levels of total and filament cytokeratins remained the same. This suggests that early estrogen exposure (i.e. 2 h) promotes interactions between cytokeratins and DNA.

A micromechanical model for three-dimensional open-cell foams is developed by using the matrix method for space frames in structural mechanics and tetrakaidcahedral unit cells. The effective elastic properties of foams are determined employing unit cells subjected to three different modes of loading. The thirty-six struts of each tetrakaidecahedral unit cell are treated as uniform slender beams, and the twenty-four vertices as rigid joints. All four-deformation mechanisms (i.e., stretching, shearing, bending and twisting) possible under the specified loadings are incorporated, and four different strut cross section shapes (i.e., circle, square, equilateral triangle and Plateau border) are treated in a unified manner. The formulas for determining the effective Young's modulus, Poisson's ratio and shear modulus of open-cell foams that are undergoing linearly elastic deformations are derived using the composite homogentization theory. The new formulas include all necessary parameters, unlike those provided by existing models.

Exposure to hypotonic stress produces a transient increase in cell volume followed by a regulatory volume decrease (RVD) in both THP-1 and HL-60 cells. In contrast, cells exposed to hypotonic stress in a high K/low Na Hanks' solution not only failed to volume regulate, but displayed a secondary swelling. Thus, while an outward K gradient was required for RVD, the secondary swelling indicated that hypotonic stress increased permeability in the absence of a negative membrane potential. The K channel blocker quinine (1-4 nM) blocked RVD in both cell types. Gramicidin's ability to overcome the quinine block of RVD indicated that RVD is mediated by a quinine-sensitive cation transport mechanism that is independent of the swelling-induced anion transport mechanism. Barium (1-4 mM), another K channel blocker, slowed the rate of RVD, while 4- aminopyridine, charybdotoxin, tetraethylammonium chloride, tetrabutylammonium chloride, and gadolinium had no effect on RVD. Furthermore, RVD was not mediated by calcium-activated conductances, since it occurred normally in Ca-free medium, in medium containing cadmium, and in BAPTA-loaded cells. Gramicidin produced little or no volume change in isotonic medium, suggesting that basal C1 permeability of both THP-1 and HL-60 cells is low. However induced an anion efflux pathway that is permeable to both chloride and bromide, but is impermeable to methanesulfonate and glutamate. The anion channel blocker 3,5- diiodosalicylic acid (DISA) antagonized RVD in both cell types. In conclusion, RVD in THP-1 and HL-60 cells is mediated by independent anion and cation transport mechanisms that involve both a DISA-sensitive anion pathway and a quinine-inhibitable K efflux pathway, neither of which requires increases in intracellular calcium to be activated.
