This article is from Anatomical Record (Hoboken, N.j. : 2007) , volume 297 . Abstract The Tasmanian devil is under threat of extinction due to the transmissible devil facial tumor disease (DFTD). This fatal tumor is an allograft that does not induce an immune response, raising questions about the activity of Tasmanian devil immune cells. T and B cell analysis has been limited by a lack of antibodies, hence the need to produce such reagents. Amino acid sequence analysis revealed that CD4, CD8, IgM, and IgG were closely related to other marsupials. Monoclonal antibodies were produced against CD4, CD8, IgM, and IgG by generating bacterial fusion proteins. These, and commercial antibodies against CD1a and CD83, identified T cells, B cells and dendritic cells by immunohistochemistry. CD4+ and CD8+ T cells were identified in pouch young thymus, adult lymph nodes, spleen, bronchus‐ and gut‐associated lymphoid tissue. Their anatomical distribution was characteristic of mammalian lymphoid tissues with more CD4+ than CD8+ cells in lymph nodes and splenic white pulp. IgM+ and IgG+ B cells were identified in adult lymph nodes, spleen, bronchus‐associated lymphoid tissue and gut‐associated lymphoid tissue, with more IgM+ than IgG+ cells. Dendritic cells were identified in lymph node, spleen and skin. This distribution is consistent with eutherian mammals and other marsupials, indicating they have the immune cell subsets for an anti‐tumor immunity. Devil facial tumor disease tumors contained more CD8+ than CD4+ cells, but in low numbers. There were also low numbers of CD1a+ and MHC class II+ cells, but no CD83+ IgM+ or IgG+ B cells, consistent with poor immune cell infiltration. Anat Rec, 297:925–938, 2014. © 2014 The Authors. Abstract Background: There is growing recognition that horizontal DNA transfer, a process known to be common in prokaryotes, is also a significant source of genomic variation in eukaryotes. Horizontal transfer of transposable elements (HTT) may be especially prevalent in eukaryotes given the inherent mobility, widespread occurrence, and prolific abundance of these elements in many eukaryotic genomes. Results: Here, we provide evidence for a new case of HTT of the transposon family OposCharlie1 (OC1) in the Tasmanian devil, Sarcophilus harrisii. Bioinformatic analyses of OC1 sequences in the Tasmanian devil genome suggest that this transposon infiltrated the common ancestor of the Dasyuridae family ~17 million years ago. This estimate is corroborated by a PCR-based screen for the presence/absence of this family in Tasmanian devils and closely-related species. Conclusions: This case of HTT is the first to be reported in dasyurids. It brings the number of animal lineages independently invaded by OC1 to 12, and adds a fourth continent to the pandemic-like pattern of invasion of this transposon. In the context of these data, we discuss the evolutionary history of this transposon family and its potential impact on the diversification of marsupials.

The Tasmanian Devil project is a cooperative effort by the Defense Modeling and Simulation Office (DMSO), Air Force Research Laboratory Warfighter Training Research Division (AFRL/HEA), and USN Air Combat Environment Test and Evaluation Facility (ACETEF) Aircraft Simulation - Manned Flight Simulator (MFS). The purpose of the project is to gain experience in the application of the HLA in the distributed mission training domain. The multi-service federation development team developed and demonstrated two simple, parallel service federations including aircraft training simulators using a common 2vX air-to-air training event. Each federation implemented a common federation design that was developed over the course of the project using the Federation Execution and Development Process (FEDEP) as a roadmap. Specific project objectives include: (1) implementation of HLA interfaces for the F-16 and F-18 simulators, an ordnance server, and a cockpit radio simulation; (2) beta testing of RTI NG; (3) first use of a VxWorks version RTI; (4) use of same FOM in two federations with two different mixes of federates; (5) use and assessment of agile FOM interfaces; and (6) examination of issues related to an evolutionary and persistent federation. This paper describes the results and lessons learned from the Tasmanian Devil project.

Abstract The Tasmanian Devil Facial Tumour Disease (DFTD) provides a unique opportunity to elucidate the long-term effects of natural and anthropogenic selection on cancer evolution. Since first observed in 1996, this transmissible cancer has caused local population declines by >90%. So far, four chromosomal DFTD variants (strains) have been described and karyotypic analyses of 253 tumours showed higher levels of tetraploidy in the oldest strain. We propose that increased ploidy in the oldest strain may have evolved in response to effects of genomic decay observed in asexually reproducing organisms. In this study, we focus on the evolutionary response of DFTD to a disease suppression trial. Tumours collected from devils subjected to the removal programme showed accelerated temporal evolution of tetraploidy compared with tumours from other populations where no increase in tetraploid tumours were observed. As ploidy significantly reduces tumour growth rate, we suggest that the disease suppression trial resulted in selection favouring slower growing tumours mediated by an increased level of tetraploidy. Our study reveals that DFTD has the capacity to rapidly respond to novel selective regimes and that disease eradication may result in novel tumour adaptations, which may further imperil the long-term survival of the world's largest carnivorous marsupial.
