30Positive Selection In Glycolysis Among Australasian Stick Insects.

This article is from BMC Evolutionary Biology , volume 13 . Abstract Background: The glycolytic pathway is central to cellular energy production. Selection on individual enzymes within glycolysis, particularly phosphoglucose isomerase (Pgi), has been associated with metabolic performance in numerous organisms. Nonetheless, how whole energy-producing pathways evolve to allow organisms to thrive in different environments and adopt new lifestyles remains little explored. The Lanceocercata radiation of Australasian stick insects includes transitions from tropical to temperate climates, lowland to alpine habitats, and winged to wingless forms. This permits a broad investigation to determine which steps within glycolysis and what sites within enzymes are the targets of positive selection. To address these questions we obtained transcript sequences from seven core glycolysis enzymes, including two Pgi paralogues, from 29 Lanceocercata species. Results: Using maximum likelihood methods a signature of positive selection was inferred in two core glycolysis enzymes. Pgi and Glyceraldehyde 3-phosphate dehydrogenase (Gaphd) genes both encode enzymes linking glycolysis to the pentose phosphate pathway. Positive selection among Pgi paralogues and orthologues predominately targets amino acids with residues exposed to the protein’s surface, where changes in physical properties may alter enzyme performance. Conclusion: Our results suggest that, for Lancerocercata stick insects, adaptation to new stressful lifestyles requires a balance between maintaining cellular energy production, efficiently exploiting different energy storage pools and compensating for stress-induced oxidative damage.

Tisamenus is endemic to the Philippine Islands and distributed throughout most of the archipelago with the exception of the Greater Mindanao PAIC, and the greatest species diversity found on the island of Luzon. Differential diagnoses as well as notes on intraspecific variability and geographic distributions, including a great number of new records, are provided for all twenty-four known species with twenty-two of them illustrated. Keys to species are presented to distinguish females and males. Seven new species are described: T. charestae Hennemann & Le Tirant n . sp . from Luzon and T. napalaki n . sp . from Luzon and Palaui are described from the female, male and egg, T. trapezoides n . sp . from Luzon from the female and male, and T. heitzmanni n . sp . from Cebu, T. irenoliti n . sp . from Marinduque, T. makinis n . sp . from Luzon and T. malawak n . sp . from Camiguin are described from the males only. Two species are found to by synonyms ( n . syn .): T. fratercula (Rehn & Rehn, 1939) is synonymised with T. cervicornis Bolívar, 1890 and T. atropos (Rehn & Rehn, 1939) is synonymised with T. clotho (Rehn & Rehn, 1939). The type-species T. serratorius Stål, 1875 is for the first time recorded from a precise locality on the island of Luzon and is re-described and illustrated along with descriptions of the previously unknown male and egg. The previously unknown males of T. armadillo Redtenbacher, 1906, T. clotho (Rehn & Rehn, 1939, T. deplanatus (Westwood, 1848), T. draconinus (Westwood, 1848) and T. hebardi (Rehn & Rehn, 1939) are described and/or illustrated for the first time, as are the previously unknown females of T. hystrix (Rehn & Rehn, 1939), T. lachesis (Rehn & Rehn, 1939), T. polillo (Rehn & Rehn, 1939) and T. spadix (Rehn & Rehn, 1939). Also, the eggs of T. armadillo Redtenbacher, 1906, T. cervicornis Bolívar, 1890, T. clotho (Rehn & Rehn, 1939), T. deplanatus (Westwood, 1848), T. draconinus (Westwood, 1848), T. hebardi (Rehn & Rehn, 1939), T. lachesis (Rehn & Rehn, 1939), T. polillo (Rehn & Rehn, 1939), T. serratorius Stål, 1875 and T. spadix (Rehn & Rehn, 1939) are described and illustrated for the first time. Tisamenus is endemic to the Philippine Islands and distributed throughout most of the archipelago with the exception of the Greater Mindanao PAIC, and the greatest species diversity found on the island of Luzon. Differential diagnoses as well as notes on intraspecific variability and geographic distributions, including a great number of new records, are provided for all twenty-four known species with twenty-two of them illustrated. Keys to species are presented to distinguish females and males. Seven new species are described: T. charestae Hennemann & Le Tirant n . sp . from Luzon and T. napalaki n . sp . from Luzon and Palaui are described from the female, male and egg, T. trapezoides n . sp . from Luzon from the female and male, and T. heitzmanni n . sp . from Cebu, T. irenoliti n . sp . from Marinduque, T. makinis n . sp . from Luzon and T. malawak n . sp . from Camiguin are described from the males only. Two species are found to by synonyms ( n . syn .): T. fratercula (Rehn & Rehn, 1939) is synonymised with T. cervicornis Bolívar, 1890 and T. atropos (Rehn & Rehn, 1939) is synonymised with T. clotho (Rehn & Rehn, 1939). The type-species T. serratorius Stål, 1875 is for the first time recorded from a precise locality on the island of Luzon and is re-described and illustrated along with descriptions of the previously unknown male and egg. The previously unknown males of T. armadillo Redtenbacher, 1906, T. clotho (Rehn & Rehn, 1939, T. deplanatus (Westwood, 1848), T. draconinus (Westwood, 1848) and T. hebardi (Rehn & Rehn, 1939) are described and/or illustrated for the first time, as are the previously unknown females of T. hystrix (Rehn & Rehn, 1939), T. lachesis (Rehn & Rehn, 1939), T. polillo (Rehn & Rehn, 1939) and T. spadix (Rehn & Rehn, 1939). Also, the eggs of T. armadillo Redtenbacher, 1906, T. cervicornis Bolívar, 1890, T. clotho (Rehn & Rehn, 1939), T. deplanatus (Westwood, 1848), T. draconinus (Westwood, 1848), T. hebardi (Rehn & Rehn, 1939), T. lachesis (Rehn & Rehn, 1939), T. polillo (Rehn & Rehn, 1939), T. serratorius Stål, 1875 and T. spadix (Rehn & Rehn, 1939) are described and illustrated for the first time. So far only nine species and two subspecies are known from the island. Two new species are described: Ramulus yasini n . sp . from both sexes and the eggs and Nescicroa bonneaui n . sp . from the female only. One new subspecies, Dimorphodes mancus morotaiense n . ssp ., is described from both sexes and the eggs. The new subspecies parallels the only known leaf insect of Morotai, Phyllium tobeloense bhaskarai Cumming et al., 2019, in representing and offshoot of the nominate Halmahera population with only the egg morphology providing concrete features that allow differentiation between the populations. Acanthoderus gravidus Bates, 1865 is removed from synonymy with Pylaemenes coronatus (Haan, 1842) and re-established as a valid species, thus becoming Pylaemenes gravidus (Bates, 1865) rev . stat . Datames aequalis Rehn, 1904 from the island of Obi is shown to be a synonym of P. gravidus ( n . syn .). The first records from the islands of Halmahera, Ternate and Morotai are range extensions for P. gravidus . Eggs of the Morotai populations of Anchiale maculata (Olivier, 1792) and Orxines xiphias (Westwood, 1859) are described and illustrated with the latter species recorded from the island of Morotai for the first time. So far only nine species and two subspecies are known from the island. Two new species are described: Ramulus yasini n . sp . from both sexes and the eggs and Nescicroa bonneaui n . sp . from the female only. One new subspecies, Dimorphodes mancus morotaiense n . ssp ., is described from both sexes and the eggs. The new subspecies parallels the only known leaf insect of Morotai, Phyllium tobeloense bhaskarai Cumming et al., 2019, in representing and offshoot of the nominate Halmahera population with only the egg morphology providing concrete features that allow differentiation between the populations. Acanthoderus gravidus Bates, 1865 is removed from synonymy with Pylaemenes coronatus (Haan, 1842) and re-established as a valid species, thus becoming Pylaemenes gravidus (Bates, 1865) rev . stat . Datames aequalis Rehn, 1904 from the island of Obi is shown to be a synonym of P. gravidus ( n . syn .). The first records from the islands of Halmahera, Ternate and Morotai are range extensions for P. gravidus . Eggs of the Morotai populations of Anchiale maculata (Olivier, 1792) and Orxines xiphias (Westwood, 1859) are described and illustrated with the latter species recorded from the island of Morotai for the first time. So far only nine species and two subspecies are known from the island. Two new species are described: Ramulus yasini n . sp . from both sexes and the eggs and Nescicroa bonneaui n . sp . from the female only. One new subspecies, Dimorphodes mancus morotaiense n . ssp ., is described from both sexes and the eggs. The new subspecies parallels the only known leaf insect of Morotai, Phyllium tobeloense bhaskarai Cumming et al., 2019, in representing and offshoot of the nominate Halmahera population with only the egg morphology providing concrete features that allow differentiation between the populations. Acanthoderus gravidus Bates, 1865 is removed from synonymy with Pylaemenes coronatus (Haan, 1842) and re-established as a valid species, thus becoming Pylaemenes gravidus (Bates, 1865) rev . stat . Datames aequalis Rehn, 1904 from the island of Obi is shown to be a synonym of P. gravidus ( n . syn .). The first records from the islands of Halmahera, Ternate and Morotai are range extensions for P. gravidus . Eggs of the Morotai populations of Anchiale maculata (Olivier, 1792) and Orxines xiphias (Westwood, 1859) are described and illustrated with the latter species recorded from the island of Morotai for the first time. Abstract Many stick insects and mantophasmids possess tarsal ‘heel pads’ (euplantulae) covered by arrays of conical, micrometre-sized hairs (acanthae). These pads are used mainly under compression; they respond to load with increasing shear resistance, and show negligible adhesion. Reflected-light microscopy in stick insects (Carausius morosus) revealed that the contact area of ‘heel pads’ changes with normal load on three hierarchical levels. First, loading brought larger areas of the convex pads into contact. Second, loading increased the density of acanthae in contact. Third, higher loads changed the shape of individual hair contacts gradually from circular (tip contact) to elongated (side contact). The resulting increase in real contact area can explain the load dependence of friction, indicating a constant shear stress between acanthae and substrate. As the euplantula contact area is negligible for small loads (similar to hard materials), but increases sharply with load (resembling soft materials), these pads show high friction coefficients despite little adhesion. This property appears essential for the pads’ use in locomotion. Several morphological characteristics of hairy friction pads are in apparent contrast to hairy pads used for adhesion, highlighting key adaptations for both pad types. Our results are relevant for the design of fibrillar structures with high friction coefficients but small adhesion.
