Abstract Aflatoxin B1 (AFB1), which causes hepatocellular carcinoma and immune-suppression, is commonly found in feedstuffs. To evaluate the ability of selenium (Se) to counteract the deleterious effects of AFB1, two hundred 1-day-old male avian broilers, divided into five groups, were fed with basal diet (control group), 0.3 mg/kg AFB1 (AFB1 group), 0.3 mg/kg AFB1+0.2 mg/kg Se (+Se group I), 0.3 mg/kg AFB1+0.4 mg/kg Se (+Se group II) and 0.3 mg/kg AFB1+0.6 mg/kg Se (+Se group III), respectively. Compared with control group, the relative weight of spleen in the AFB1 group was decreased at 21 days of age. The relative weight of spleen in the three +Se groups was higher than that in the AFB1 group. By pathological observation, the major spleen lesions included congestion in red pulp and vacuoles appeared in the lymphatic nodules and periarterial lymphatic sheath in the AFB1 group. In +Se groups II and III, the incidence of major splenic lesions was decreased. The percentages of CD3+, CD3+CD4+ and CD3+CD8+ T cells in the AFB1 group were lower than those in control group from 7 to 21 days of age, while there was a marked increase in the three +Se groups compared to the AFB1 group. The results indicated that sodium selenite could improve the cellular immune function impaired by AFB1 through increasing the relative weight of spleen and percentages of splenic T cell subsets, and alleviating histopathological spleen damage.

Abstract Nano-selenium (Se), with its high bioavailability and low toxicity, has attracted wide attention for its potential application in the prevention of oxidative damage in animal tissues. However, the effect of nano-Se of different sizes on the intestinal epithelial cells of the crucian carp (Carassius auratus gibelio) is poorly understood. Our study showed that different sizes and doses of nano-Se have varied effects on the cellular protein contents and the enzyme activities of secreted lactate dehydrogenase, intracellular sodium potassium adenosine triphosphatase, glutathione peroxidase, and superoxide dismutase. It was also indicated that nano-Se had a size-dependent effect on the primary intestinal epithelial cells of the crucian carp. Thus, these findings may bring us a step closer to understanding the size effect and the bioavailability of nano-Se on the intestinal tract of the crucian carp.

Abstract Determining the speciation of selenium in vivo is crucial to understanding the biological activity of this essential element, which is a popular dietary supplement due to its anti-cancer properties. Hyphenated techniques that combine separation and detection methods are traditionally and effectively used in selenium speciation analysis, but require extensive sample preparation that may affect speciation. Synchrotron-based X-ray absorption and fluorescence techniques offer an alternative approach to selenium speciation analysis that requires minimal sample preparation. We present a brief summary of some key HPLC-ICP-MS and ESI-MS/MS studies of the speciation of selenium in cells and rat tissues. We review the results of a top-down approach to selenium speciation in human lung cancer cells that aims to link the speciation and distribution of selenium to its biological activity using a combination of X-ray absorption spectroscopy (XAS) and X-ray fluorescence microscopy (XFM). The results of this approach highlight the distinct fates of selenomethionine, methylselenocysteine and selenite in terms of their speciation and distribution within cells: organic selenium metabolites were widely distributed throughout the cells, whereas inorganic selenium metabolites were compartmentalized and associated with copper. New data from the XFM mapping of electrophoretically-separated cell lysates show the distribution of selenium in the proteins of selenomethionine-treated cells. Future applications of this top-down approach are discussed.

The proposed study required the creation of prostate cell lines that contain hSP56 transgene and it is regulated artificially. During the first year of this study, preparation of inducible mammalian expression construct of hSP56 gene and anti sense expression construct was accomplished. Prostate cancer cell line PC-3 expresses hSP56 extremely low and they are stably transfected with the above-described expression constructs. Independent clones of PC-3 cells that contain transgene of hSP56 were selected and screened for inducible expression of hSP56 by immunoblotting. Anti-sense R&A method was used to down-regulated the endogenous hSP56 in LNCaP cells and an effective interference RNA was identified for hSP56 gene. The down regulation of hSP56 in LNCaP cells was confirmed by immunoblotting as well.

Come Rack! Come Rope! is a historical novel by the English priest and writer Robert Hugh Benson, a convert to Catholicism from Anglicanism. Set in Derbyshire at the time of the Elizabethan persecution of Catholics, when being or harboring a priest was considered treason and was punishable with death, it tells the story of two young lovers who give up their chance of happiness together, choosing instead to face imprisonment and martyrdom, so that "God's will" may be done. <br><br>The book was written nearly nine years after Benson's reception into the Catholic Church. The inspiration for the story comes from Dom Bede Camm's account of the recusant Fitzherbert family in Forgotten Shrines (1910), and from Benson's own visit in 1911 to Padley, home of the Fitzherberts, and scene of part of the novel, in order to preach at the annual pilgrimage there. The title of the book is taken from a letter of Saint Edmund Campion in which, after torture, he assured Catholics that he had revealed "no things of secret, nor would he, come rack, come rope." Benson was was a prolific and popular writer during his time, and in 1903 he became a prominent convert to the Roman Catholic Church from Anglicanism . Robert Hugh Benson, who was educated at Eton College and Trinity College, Cambridge, was ordained an Anglican priest in 1895 by his father, the Archbishop of Canterbury. Eight years later, after weighty consideration, Robert Benson converted to Roman Catholicism. In 1904 he was ordained a Roman Catholic priest and took up residence in Cambridge, where he ministered to Roman Catholic students as their unofficial chaplain. In 1911, he was made a monsignor. In a former book, called "Lord of the World," I attempted to sketch the kind of developments a hundred years hence which, I thought, might reasonably be expected if the present lines of what is called "modern thought" were only prolonged far enough; and I was informed repeatedly that the effect of the book was exceedingly depressing and discouraging to optimistic Christians. In the present book I am attempting -- also in parable form -- not in the least to withdraw anything that I said in the former, but to follow up the other lines instead, and to sketch -- again in parable -- the kind of developments, about sixty years hence which, I think, may reasonably be expected should the opposite process begin, and ancient thought (which has stood the test of centuries, and is, in a very remarkable manner, being "rediscovered" by persons even more modern than modernists) be prolonged instead.<br><br>We are told occasionally by moralists that we live in very critical times, by which they mean that they are not sure whether their own side will win or not. In that sense no times can ever be critical to Catholics, since Catholics are never in any kind of doubt as to whether or no their side will win. But from another point of view every period is a critical period, since every period has within itself the conflict of two irreconcilable forces. It has been for the sake of tracing out the kind of effects that, it seemed to me, each side would experience in turn, should the other, at any rate for a while, become dominant, that I have written these two books.
