7The Effect Of Amino Acid L-Phenylalanine, Selenium And Nitroxin Biological Fertilizer On The Physicochemical Properties Of Goji Berries (Lycium Barbarum Cv. GB1)

Introduction The Lycium genus of the Solanaceae family has excellent nutritional and medicinal value. Two species of Lycium barbarum L. and Lycium chinense Mill. It is often called wolfberry or goji berry. The use of amino acids for horticultural crops is common worldwide, and many chemicals used as biostimulants are mixtures of amino acids. The effect of amino acids on plants seems to depend on the type of amino acid supplied and the type of plant. Selenium (Se) is an important component of selenoproteins and seleno-amino acids. Therefore, it has played many roles in the growth and function of living cells and important biological functions in animals and humans. Also Se is very similar in properties to sulfur and can act as S in biochemical systems. Biological fertilizers are fertile materials that contain one or more beneficial soil organisms within a suitable carrier. Overall, these fertilizers contain different types of microorganisms that can convert nutrients from unavailable form to available form through a biological process. The application of supernitroxin fertilizer by stimulating the synthesis of plant hormones increased the growth indicators of sesame varieties.   Materials and Methods This experiment was conducted at the Research farm University of Mashhad, during 2021 and 2022 years. In early May, goji berry seedlings were planted in the field to check the effect of the L-phenylalanine (Phe), sodium selenate (Se), and nitroxine were applied before harvesting and foliar spraying on the goji berry plant during the growth stages. To conduct experiment, two-year-old seedlings of Goji berry cultivar GB1 were obtained from Mashhad Seedling Company located in Razavi Khorasan province, and after the seedlings were transferred to ground and established, initial foliar spraying was done. In order to evaluate the effect of L-phenylalanine, selenium, and nitroxine treatments, experiment were conducted as a randomized complete block design in 5 replications which factors include the amino acid L-phenylalanine (Phe: 0.5, 1, and 1.5 mM), sodium selenate (Se: 0.25, 0.5, and 1 mg.L -1 ) and nitroxin biological fertilizer (170, 330, and 500 μl.L -1 ) at three levels and distilled water was applied as a control. One pot was considered for each repetition and a total of 10 pots were considered along with the control. Plants were sprayed every 15 days after establishment. After the three stages of foliar spraying, the content of phenol and flavonoid content and antioxidant activity, PAL and anthocyanin were measured.   Results and Discussion The results showed that the treatments used in this experiment had a significant effect on the physiological and chemical characteristics of the goji berry plant. The results showed that the highest amount of titratable acidity (0.896%) was obtained in samples treated with nitroxine at a concentration of 333 microliters. The highest amount (18.56 mg.g -1 ) of this index was obtained in fruits treated with phenylalanine at a concentration of 1.5 mM. The highest amount of this index was obtained in the fruits treated with phenylalanine at a concentration of 1 mM, which was 61.98% higher than the amount of flavonoid recorded in goji berries under control conditions. Also, the results showed that despite the decrease in the activity of PAL enzyme during the increase of selenium concentration, the activity of this enzyme was 13.66% higher than the activity of PAL enzyme under the condition of using selenium at a concentration of 1 mg/liter. The increase in the functioning of the antioxidant system is determined by the total antioxidant, which is controlled by the content of low-molecular components and the activity of antioxidant enzymes. Compounds such as ascorbic acid, glutathione, tocopherol, carotenoids, anthocyanins, endogenous metal chelators, TPC, TFC and alkaloids are low molecular weight antioxidants. Also nitroxin, supernitroplus, phosphate and mycorrhizal fertilizers. Nitroxin binds N in the atmosphere and balances the absorption of nutrient element in the plant. Nitroxin is responsible for the secretion of amino acids, antibiotics, hydrogen cyanide and siderophores, which promote the growth and development of plant roots and shoots, protect the roots from pathogens, thereby increasing yield. A sufficient supply of phenylalanine through the use of exogenous Phe or endogenous Phe provided by the shikimic acid pathway may be required to stimulate the activity of the phenylpropanoid pathway, as shown by the high PAL activity responsible for the accumulation of phenols, flavocyanins and anthocyanins. This leads to the need for ROS accumulation. Anthocyanins are groups of flavonoids found in vacuoles epidermal and mesophyll cells of plants. Anthocyanins can protect chlorophyll from light oxidation and, compared to other components, is a better indicator of plant oxidative stress caused by external factors that accumulate in plants.   Conclusions The present study was conducted with the aim of studying the effect of L-phenylalanine, selenium and nitroxin biofertilizer on improving the vegetative growth, yield and secondary metabolites of gojiberry during two cropping years. To conduct the experiment, two-year-old seedlings of Goji berry variety GB1 were obtained from Mashhad Seedling Company located in Razavi Khorasan province. The results demonstrated that the application of L-phenylalanine amino acid, selenium, and nitroxin biofertilizer significantly influenced the traits studied during the experiment. Specifically, nitroxin at a concentration of 166 microliters increased the amount of soluble solids, while at a concentration of 333 microliters, it elevated titratable acidity compared to the control samples. The highest PAL enzyme activity was observed with phenylalanine at a concentration of 1.5 mM, showing a 52.17% increase compared to the PAL enzyme activity under control conditions.

The purpose of this study is to evaluate existing manual methods for analyzing asbestos, beryllium, lead, cadmium, selenium, and mercury, and from this evaluation to provide the best and most practical set of analytical methods for measuring emissions of these elements from stationary sources. The work in this study was divided into two phases. Phase I was limited to surveying sources of information and summarizing the findings in terms of existing methods; totally new methods were not developed. However, in the case of asbestos and mercury, it was necessary to modify existing methods significantly, and some laboratory and field testing was performed during Phase I to develop these two methods. Phase II was concerned with the testing, evaluation, and modification of the methods of analysis developed during Phase I. Both laboratory and field tests were conducted during this phase of the work. (Author/BT)

Introduction [1] : Selenium is an essential trace element, which plays a key role in the development of reproductive performance and animal safety. This mineral is an important part of at least 25 effective proteins in a variety of physiological functions, including increased resistance to oxidative stress, DNA repair, and improvement to reproductive performance and immune function. Despite the fact that selenium requirement of birds can be met by corn-soybean meal diet, dietary sodium selenite up to a maximum level of 0.5 mg kg -1 is highly recommended to improve avian health and productive performance. However, interaction with other minerals, poor retention and lower deposition efficiency questioning the incorporation of sodium selenite into the diet. Vitamin E is the main antioxidant in egg yolk lipid terminating the lipid peroxidation chain by reacting with lipid peroxides and making them be stable. Diet supplementation with vitamin E could improve performance and egg quality and provided health benefits to laying hens. The positive effects of Se and vitamin E on egg production, egg quality traits has been extensively studied in laying hens, but information regarding the potential benefits of these antioxidants on the productive performance of Japanese quails is limited in literature. Thus, this study was conducted to investigate the combined effects of various sources of selenium (0.4 mg kg-1) plus vitamin E (120 mg kg-1) on performance, qualitative characteristics, immune system and blood metabolites of Japanese quails.   Materials and Methods: A total of 144 12-week-old female Japanese quails were randomly divided into three treatment groups with four replicates and twelve birds in each in a completely randomized design. The experimental treatments consisted of basal diet (with no supplementation) and two organic (sel-plex) and inorganic (sodium selenite) sources of selenium which provided 0.4 mg/kg, plus 120 mg of vitamin E per kilogram of diet. Feed intake, feed conversion ratio (FCR), egg production and quality parameters in fresh and stored eggs, cell immunity, blood cholesterol, triglyceride and total antioxidant contents were evaluated. At the end of experiment, egg yolk selenium and malondialdehyde concentrations were measured. The thiobarbituric acid reactive substances (TBARS) assay with small modification was used to measure lipid oxidation. Briefly, 2 grams of homogenous yolk was mixed with 5 mL of 20 % trichloroacetic acid (TCA) and 4 mL distilled water, and homogenized for 30 s at high speed. The homogenate was then centrifuged (1000 g for 20 min) and the resulting supernatant was filtered with Whatman Filter Paper (grade no. 1). Two mL of filtrated was mixed with 2 mL of thiobarbituric acid (TBA, 0.02 M) in a test tube and heated in boiling water for 20 min. After cooling, the absorbance of the resulting solution was read with a spectrophotometer at 532 nm.   Results and discussion: The results showed that egg weight, egg laying percentage and feed conversion rate were not affected by selenium organic and inorganic sources compared to control group (P > 0.05) . Birds receiving organic selenium along with vitamin E in their diets had low feed intake compared to other treatments and their feed conversion ratio numerically decreased and intended to significant (p<0.1).The organic selenium plus vitamin E improved the height of white, HU units, yolk pH, and yolk index compared to control group. Egg quality parameters including yolk index and shell surf index in inorganic selenium group were significantly higher than control group (p<0.0.05). Egg yolk selenium was increased in organic and organic selenium (2.06, 1.31 mg/kg) compared to control (0.36), which is a better value from the nutritional viewpoint. Blood cholesterol and triglyceride levels were not affected by experimental diets. Concentration of yolk MDA was highest in control and lowest in organic selenium group during storage (P < 0.05). The result of this study showed that egg production, feed conversion ratio and total egg mass were not affected by experimental treatments. This was also observed by other researchers who evaluated the dietary inclusion of vitamin E. In another one egg quality parameters, however, egg weight was significantly raised after being fed with a diet contained 120 mg kg -1 of vitamin E and 0.4 mg kg -1 of organic selenium.   Conclusion: In conclusion, our study data revealed dietary organic selenium and vitamin E, could improve the egg quality during storage and increased egg yolk selenium of Japanese quails.

Introduction: [1] Selenium is an essential mineral for many metabolic functions of the body, including the activation of enzymes and the optimal biochemical and physiological function of birds ( Antongiovanni et al ., 2007 ). Selenium protects cell membranes from oxidative damage and can therefore improve nutrient efficiency ( Arner et al., 2012 ). The small intestine is the most important part in the digestion and absorption of nutrients, while the large intestine and intestinal tract are very important areas for the accumulation of microbes ( Chitra et al ., 2013 ). Gastrointestinal health is one of the most important and effective factors in bird function. Gastrointestinal microbial population affects the nutrition and health of various animal species, including poultry. These microorganisms need trace elements such as selenium to perform their normal metabolic functions. Selenium may affect bacterial cells by disrupting the respiratory chain ( Pappas et al ., 2005 ). In addition to improving the quality and composition of intestinal microflora, selenium can have a positive effect on the morphology of the intestine as an antioxidant ( Haghighi-khoshkhoo et al ., 2010 ). Despite these benefits, the effect of selenium on intestinal microbial population is largely unknown, so the aim of this study was to investigate the effect of different levels of organic and inorganic selenium in the diet on microbial population, intestinal morphology and intestinal acidity in laying hens. Materials and Methods: The experiment was conducted in a completely randomized design with 300 laying hens of high-line strains from 23 to 35 weeks of age with 5 treatments, 6 replications and 10 hens per replication. Experimental treatments include 1- Base diet (without selenium), 2- Base diet + 0.5 mg/kg sodium - selenite, 3- Base diet + mg/kg 1 selenite - sodium, 4- Base diet + mg/kg 0.5 selenium-methionine, 5-base diet + 1 mg/kg selenium-methionine. At the end of the experiment, two birds were randomly selected from each replicate; To evaluate the microbial population, a sample of the contents of the cecum on the culture medium was used (1). Tissue samples were prepared and then measured using a microscope, villi length, villi width, crypt depth, and number of goblet cells (9). To measure acidity, samples were taken from the contents of the cecum, and acidity was measured by a pH meter. Results and Discussion: The results indicate that consuming 1 mg/kg of selenium-methionine increased the villi area compared to the control treatment. The experimental treatments did not affect villi length, villi width, crypt depth, number of goblet cells, and the ratio of villus length to crypt depth. The consumption of 1 mg/kg of selenium-methionine significantly decreased the population of Salmonella and increased the population of Lactobacillus in the cecal compared to other experimental treatments. The consumption of 0.5 and 1 mg/kg of selenium-methionine caused a significant decrease in the population of aerobic bacteria compared to the control treatment. The acidity of cecal contents in the treatment containing 1 mg/kg of selenium-methionine was significantly reduced compared to the control treatment. Several studies ( Langhout et al ., 1999 , Lukaszewicz et al ., 2011 , Pappas et al ., 2005 ) have reported that using organic sources of selenium reduces the coliform population. In mice, dogs, and laying hens, selenium intake has been shown to increase the number of Lactobacillus and decrease Escherichia coli and Staphylococcus aureus in the cecum ( Langhout et al ., 1999 , Lukaszewicz et al ., 2011 , Pappas et al. , 2005 ). Increasing the population of beneficial bacteria due to the provision of sufficient selenium for their synthesis is also an antioxidant property of selenium in preserving the life of these bacteria. It appears that beneficial bacteria, such as Lactobacillus, can competitively eliminate harmful bacteria such as Escherichia coli in the gut ( Lukaszewicz et al ., 2011 ). An increase in the number of pathogenic bacteria in the intestine causes the villi to shorten and the lining to shrink ( Attia et al ., 2010 ).Numerous studies demonstrate that the consumption of diets containing selenium compounds has destructive effects on harmful intestinal bacteria ( Chantiratikul et al ., 2008 , Hashemi et al ., 2012 , Heindl et al. , 2010 , Horn et al ., 2009 , Langhout et al ., 1999 ). Adding organic selenium to the diet of broilers increases the weight of the intestines due to the growth of villi and intestinal lamina propria ( Haghighi-khoshkhoo et al ., 201 0 , Heindl et al ., 2010 ). The lack of selenium consumption on morphology can be attributed to the levels of selenium used, as well as the bird's lack of stress. Selenium can exert its effect more effectively under stress conditions. Most harmful bacteria grow in an environment with acidity close to 7 or slightly higher, while beneficial bacteria multiply in an acidic environment and compete with pathogenic bacteria ( Cooke et al . 1973 ). An increase in acidity leads to a decrease in Escherichia coli and Salmonella in the gastrointestinal tract. Therefore, the consumption of organic sources of selenium reduces the population of pathogenic microorganisms (aerobic bacteria, Escherichia coli, and Salmonella) and increases the population of Lactobacillus competitively, followed by an increase in gastrointestinal acidity. This improvement may lead to an increase in digestion, absorption, and performance ( Kasaikina et al ., 2011 ). Conclusion:   According to the results of this study, it can be said that consuming organic sources of selenium (levels of one and 0.5 mg of selenium-methionine per kg of feed) significantly reduces the population of harmful aerobic bacteria, bacteria. Salmonella and Escherichia coli and cause a competitive increase in the beneficial population of Lactobacillus; The consequence of this operation is to increase the acidity of the cecum and reduce the damage and microbial destruction to the intestinal tissue, and therefore with positive changes in the morphology of the small intestine will lead to improved bird function.

This article is from Archives of Medical Science : AMS , volume 8 . Abstract Introduction: Selenium and zinc are indispensable microelements for normal functioning and development of the human body. They are cofactors of many enzymes of the antioxidative barrier (selenium – glutathione peroxidase; zinc – superoxide dismutase). The aim of the study was to evaluate the importance of selenium and zinc in the pathogenesis of food allergy in small children. Material and methods: The study was performed in 134 children with food allergy, aged 1 to 36 months. The control group was composed of 36 children at the same age, without clinical symptoms of food intolerance. Each child had estimated serum levels of zinc and selenium. Furthermore, the authors evaluated activity of glutathione peroxidase (GSH-Px) in erythrocyte lysates and serum. Tests were performed twice, before and after 6-month administration of elimination diet. Results: The obtained results showed that children with food allergy had significantly lower concentrations of selenium, zinc and examined enzymes in comparison to children from the control group. Concentration of selenium and zinc as well as activity of examined enzymes increased after application of eliminative diet. Conclusions: In children with allergy decreased concentrations of selenium and zinc, and lower values of glutathione peroxidase and superoxide dismutase which increased after elimination diet were affirmed. These observations suggest their role in pathogenesis of food allergy. Conducted observations indicate the need to monitor trace elements content in the diet in children with food allergy. The results showed that children with food allergy had a weakened antioxidative barrier.

Abstract Background: Objectives were to examine the effects of selenium (Se) supply and maternal nutritional plane during gestation on mammary gland growth, cellular proliferation, and vascularity at parturition and d 20 of lactation. Rambouillet primiparous ewes (n = 84) were allocated to treatments in a 2 x 3 factorial. Factors were dietary Se (adequate Se [ASe, 11.5 μg/kg BW] or high Se [HSe, 77.0 μg/kg BW]) and nutritional plane (60% [RES], 100% [CON], or 140% [EXC]). At parturition, lambs were removed and 42 ewes (7/treatment) were necropsied. Remaining ewes were fed a common diet meeting requirements for lactation and mechanically milked twice daily until necropsy on d 20. At both necropsy periods, mammary glands were dissected and tissues harvested. Samples were analyzed for RNA, DNA, and protein content, cell proliferation, and vascularity. Where interactions were present (P ≤ 0.05), least squares means from the highest-order interaction are presented. Results: Final body weight of ewes was least (P ≤ 0.002) in RES, intermediate for CON, and greatest for EXC, regardless of stage of the ewe at necropsy (parturition or d 20 of lactation). In ewes necropsied at parturition, mammary glands were heavier (P = 0.02) in EXC compared to RES, with CON intermediate. Concentration of RNA (mg/g) was decreased (P = 0.01) in EXC compared to CON at parturition. There was a tendency (P = 0.07) for a Se by nutrition interaction in percentage of cells proliferating where ASe-EXC ewes had greater (P ≤ 0.02) number of proliferating cells then all other treatments. Mammary vascular area tended (P = 0.08) to be affected by a Se by nutrition interaction where ASe-CON had less (P = 0.007) vascular area than HSe-CON ewes. In ewes necropsied at d 20 of lactation, the number of alveoli per area was decreased (P ≤ 0.05) in RES compared to CON and EXC-fed ewes. Conclusions: Results of this study indicate that proper maternal nutritional plane during gestation is important for mammary gland development, even out to d 20 of lactation.

Nutritional biofortification of foods is a promising alternative to reduce Selenium deficiency in the diet of populations. This study evaluated the biofortification capacity of jambu with Se. The experiment was completely randomized with six treatments and five repetitions, in hydroponics. Five doses of Se in the form of sodium selenate (1, 2, 3, 4 and 5 mg.L-1) and the control dose were used. Biometric, macro and micronutrient analyzes were performed, as well as the Se content in the plant parts. The indices of translocation (TrI), tolerance (tI) and Nutrient Use Efficiency (Nue) were estimated. The results were submitted to ANOVA and principal components analysis for the construction of multivariate indicators, in order to specify the regression models. The plant obtained higher agronomic performance when submitted to a dose of 3mg.L-1, with Se translocation above 70%. The tI and Nue indices indicated that jambu reached optimal growth with the dose of 3 mg.L-1 of Se. The results obtained from the regression equation of the multivariate indicators of growth, mass and nutrition indicated that the ideal concentrations of Se varied between 2.77 and 3.36 mg.L-1. The general indicator that captured the entire plant behavior showed that the optimal concentration for biofortification is 2.98 mg.L-1 of Se. The daily consumption of 100 g of biofortified jambu at the indicated dose provides a daily content of 50.13µg of Se for the population, a sufficient amount of Se for a balanced diet.

49Assessing Plasma Levels Of Selenium, Copper, Iron And Zinc In Patients Of Parkinson's Disease.

This article is from PLoS ONE , volume 8 . Abstract Trace elements have been recognized to play an important role in the development of Parkinson’s disease (PD). However, it is difficult to precisely identify the relationship between these elements and the progression of PD because of an insufficient number of patients. In this study, quantifications of selenium (Se), copper (Cu), iron (Fe) and zinc (Zn) by atomic absorption spectrophotometry were performed in plasma from 238 PD patients and 302 controls recruited from eastern China, which is so far the largest cohort of PD patients and controls for measuring plasma levels of these elements. We found that plasma Se and Fe concentrations were significantly increased whereas Cu and Zn concentrations decreased in PD patients as compared with controls. Meanwhile, these four elements displayed differential changes with regard to age. Linear and logistic regression analyses revealed that both Fe and Zn were negatively correlated with age in PD patients. Association analysis suggests that lower plasma Se and Fe levels may reduce the risk for PD, whereas lower plasma Zn is probably a PD risk factor. Finally, a model was generated to predict PD patients based on the plasma concentrations of these four trace elements as well as other features such as sex and age, which achieved an accuracy of 80.97±1.34% using 10-fold cross-validation. In summary, our data provide new insights into the roles of Se, Cu, Fe and Zn in PD progression.

Abstract Aflatoxin B1 (AFB1), which causes hepatocellular carcinoma and immune-suppression, is commonly found in feedstuffs. To evaluate the ability of selenium (Se) to counteract the deleterious effects of AFB1, two hundred 1-day-old male avian broilers, divided into five groups, were fed with basal diet (control group), 0.3 mg/kg AFB1 (AFB1 group), 0.3 mg/kg AFB1+0.2 mg/kg Se (+Se group I), 0.3 mg/kg AFB1+0.4 mg/kg Se (+Se group II) and 0.3 mg/kg AFB1+0.6 mg/kg Se (+Se group III), respectively. Compared with control group, the relative weight of spleen in the AFB1 group was decreased at 21 days of age. The relative weight of spleen in the three +Se groups was higher than that in the AFB1 group. By pathological observation, the major spleen lesions included congestion in red pulp and vacuoles appeared in the lymphatic nodules and periarterial lymphatic sheath in the AFB1 group. In +Se groups II and III, the incidence of major splenic lesions was decreased. The percentages of CD3+, CD3+CD4+ and CD3+CD8+ T cells in the AFB1 group were lower than those in control group from 7 to 21 days of age, while there was a marked increase in the three +Se groups compared to the AFB1 group. The results indicated that sodium selenite could improve the cellular immune function impaired by AFB1 through increasing the relative weight of spleen and percentages of splenic T cell subsets, and alleviating histopathological spleen damage.
