Nanotechnology has built its application in numerous fields because of its high efficiency. It is an emerging field where novel ideas are being brought to make our life easier and better. Selenium is a micronutrient and it has a vital role in the metabolism of humans and other organisms. Its absorption and efficiency increase when they are in the form of nanoparticles. This work is about the synthesis of Selenium nanoparticles from sweet potatoes and coconut water by using the bio-synthesis method and coating them on cotton fabrics to treat mastitis. After the reduction of selenite, brick red coloured powder was observed. The UV spectrometry analysis was used to indicate the presence of Selenium. In biosynthesis, functional groups will remain in these nanoparticles. The morphological features of Selenium nanoparticles were studied using XRD, SEM and EDAX. The antioxidative and antimicrobial studies of these nanoparticles were examined.

Se is an essential nutrient in the mammalian diet that has some anti-inflammatory properties and, at sufficient amounts in the diet, has been shown to be protective in various inflammatory-based disease models. More recently, Se has been shown to alter the expression of eicosanoids that orchestrate the initiation, magnitude and resolution of inflammation. Many of the health benefits of Se are thought to be due to antioxidant and redox-regulating properties of certain selenoproteins. The present review will discuss the existing evidence that supports the concept that optimal Se intake can mitigate dysfunctional inflammatory responses, in part, through the regulation of eicosanoid metabolism. The ability of selenoproteins to alter the biosynthesis of eicosanoids by reducing oxidative stress and/or by modifying redox-regulated signalling pathways also will be discussed. Based on the current literature, however, it is clear that more research is necessary to uncover the specific beneficial mechanisms behind the anti-inflammatory properties of selenoproteins and other Se metabolites, especially as related to eicosanoid biosynthesis. A better understanding of the mechanisms involved in Se-mediated regulation of host inflammatory responses may lead to the development of dietary intervention strategies that take optimal advantage of its biological potency.

This article is from Critical Care , volume 18 . Abstract Introduction: Low plasma selenium concentrations are frequent in critically ill patients. However, whether this is due to systemic inflammation, a deficient nutritional state or both is still not clear. We aimed to determine the factors associated with low plasma selenium in critically ill children while considering the inflammatory response and nutritional status. Method: A prospective study was conducted in 173 children (median age 34 months) with systemic inflammatory response who had plasma selenium concentrations assessed 48 hours after admission and on the 5th day of ICU stay. The normal reference range was 0.58 μmol/L to 1.6 μmol/L. The outcome variable was ‘low plasma selenium’, which was defined as plasma selenium values below the distribution median during this period. The main explanatory variables were age, malnutrition, sepsis, C-reactive protein (CRP), and clinical severity scores. The data were analyzed using a Binomial Generalized Estimating Equations model, which includes the correlation between admission and 5th day responses. Results: Malnutrition and CRP were associated with low plasma selenium. The interaction effect between these two variables was significant. When CRP values were less than or equal to 40 mg/L, malnutrition was associated with low plasma selenium levels (odds ratio (OR) = 3.25, 95% confidence interval (CI) 1.39 to 7.63, P = 0.007; OR = 2.98, 95% CI 1.26 to 7.06, P = 0.013; OR = 2.49, 95% CI 1.01 to 6.17, P = 0.049, for CRP = 10, 20 and 40 mg/L, respectively). This effect decreased as CRP concentrations increased and there was loose significance when CRP values were >40 mg/L. Abstract Lysine methylation has been extensively studied in histones, where it has been shown to provide specific epigenetic marks for the regulation of gene expression; however, the molecular mechanism and physiological function of lysine methylation in proteins other than histones remains to be fully addressed. To better understand the substrate diversity of lysine methylation, S-adenosylmethionine (SAM) derivatives with alkyne-moieties have been synthesized. A selenium-based SAM analog, propargylic Se-adenosyl-l-selenomethionine (ProSeAM), has a wide spectrum of reactivity against various lysine methyltransferases (KMTs) with sufficient stability to support enzymatic reactions in vitro. By using ProSeAM as a chemical probe for lysine methylation, we identified substrates for two seven-beta-strand KMTs, METTL21A and METTL10, on a proteomic scale in mammalian cells. METTL21A has been characterized as a heat shock protein (HSP)-70 methyltransferase. Mammalian METTL10 remains functionally uncharacterized, although its ortholog in yeast, See1, has been shown to methylate the translation elongation factor eEF1A. By using ProSeAM-mediated alkylation followed by purification and quantitative MS analysis, we confirmed that METTL21A labels HSP70 family proteins. Furthermore, we demonstrated that METTL10 also methylates the eukaryotic elongation factor EF1A1 in mammalian cells. Subsequent biochemical characterization revealed that METTL10 specifically trimethylates EF1A1 at lysine 318 and that siRNA-mediated knockdown of METTL10 decreases EF1A1 methylation levels in vivo. Thus, our study emphasizes the utility of the synthetic cofactor ProSeAM as a chemical probe for the identification of non-histone substrates of KMTs.

This article is from Nutrients , volume 1 . Abstract Requirements for trace minerals in camels, particularly selenium, are not well-known. Selenium supplementation using a pharmaceutical form or commercial mineral mixture is common practice in camels to address the cardiomyopathy often attributed to selenium deficiency. This supplementation is often empirical and based on estimated needs for cattle. Nowadays the use of selenium in animal foodstuffs is commonplace and further investigation of its metabolism (ingestion, dynamic of storage-destocking, excretion) in camels is warranted. The present review aimed to synthesize all the experimental research (comparative selenium status in cow and camel, response to different levels of supplementation at different physiological stages, excretion maternal transfer, experimental toxicosis) and field observations (deficiency, supplementation practices) undertaken in camels. The results underline the particularity of the unique metabolic profile of the camel and lead to practical recommendations for supplementation in camels, highlighting its relative sensitivity to excess Se intake at lower levels than in cattle. The maximal tolerable dose is 8 mg and the recommended doses range from 2 to 4 mg.

A large body of data suggests that selenium supplementation may be used as a chemopreventative strategy to reduce the risk of prostate cancer. In spite of this, little is known about the use of selenium as a cancer therapy. The inorganic form of selenium, selenite, undergoes thiol-dependent reduction to selenide, which supplies selenium for the synthesis of selenoproteins. At lower concentrations, the major effects of selenite are related to its role as a micronutrient. However, at higher concentrations, selenite may become toxic. Excessive selenite metabolism depletes cells of the primary intracellular antioxidant, glutathione, and generates superoxide. The net effect of the metabolism of selenite is a profound alteration in the cellular redox status and generation of potentially lethal reactive oxygen species. The authors characterized the response of androgen-dependent LAPC-4 prostate cancer cells, as well as patient-matched pairs of normal and malignant prostate cells to selenite. Selenite-induced growth inhibition and apoptosis were correlated with changes in Bcl-2 family member expression, altered intracellular GSH status, and MnSOD expression. They also examined the ability of selenite to sensitize prostate cancer cells to gamma irradiation. The primary goal of this proposal was to generate preclinical data supporting the concept that selenite might be useful as a novel chemotherapeutic agent alone or in combination with radiation therapy to treat prostate cancer.

The hypothesis of selenium chemoprevention is principally formulated by the observations that cancer incidence is inversely associated with selenium status. However, recent clinical and epidemiological studies demonstrate a role for some selenoproteins in exacerbating or promoting other disease states, specifically type 2 diabetes, although other data support a role of selenium in stimulating insulin sensitivity. Therefore, it is clear that our understanding in the role of selenium in glucose metabolism and chemoprevention is inadequate and incomplete. Research exploring the role of selenium in individual healthcare is of upmost importance and possibly will help explain how selenium is a double-edged sword in the pathologies of chronic diseases.

Skeletal damage may be the critical result of low-level prolonged exposure to several xenobiotics in the general population, but the mechanisms of their adverse effects are not clearly understood. The current study was aimed to investigate the possible ability of simultaneous subchronic peroral administration of selenium (Se) and diazinon (DZN) to induce changes in bone of adult male rats.In our study, twenty 1-month-old male Wistar rats were randomly divided into two experimental groups. In the first group, young males were exposed to 5 mg Na2SeO3/L and 40 mg of DZN/L in drinking water, for 90 days. Ten 1-month-old males without Se and DZN intoxication served as a control group. At the end of the experiment, macroscopic and microscopic structures of the femurs were analysed using analytical scales, sliding instrument, and polarized light microscopy. Methods:: In a case-control study, 43 newly diagnosed pemphigus vulgaris patients were compared with 58 healthy people from 2009 to 2010. The severity of the disease was estimated according to Harman’s scores. Serum selenium was measured with atomic absorption but serum zinc and copper concentrations were determined spectrophotometrically. Data were compared with independent t test. Correlations were evaluated by Pearson correlation test. Results:: Both groups were the same based on sex, age, and weight and body mass index. The mean duration of disease was 5.6 month. Selenium (Se) is an essential trace element, and its deficiency in the humans leads to increase the risk of various diseases, such as cancer and heart diseases. The objective of this study was to investigate the influence of biofield energy treatment on the physical and thermal properties of the selenium powder. The selenium powder was divided into two parts denoted as control and treated. The Control part was remained as untreated and treated part received Mr. Trivedi’s biofield energy treatment. Both control and treated selenium samples were characterized using x-ray diffraction (XRD), differential scanning calorimetry (DSC), thermogravimetric analysis – differential thermal analysis (TGA-DTA), and Fourier transform infrared spectroscopy (FT-IR). The XRD data showed that biofield energy treatment has slightly altered the lattice parameter (0.07%), unit cell volume (0.15%), density (-0.14%), atomic weight (0.15%), and nuclear charge per unit volume (-0.21%) in the treated selenium powder as compared to the control. The crystallite size of the treated selenium powder was reduced considerably from 106.98 nm (control) to 47.55 nm. The thermal analysis study showed that the latent heat of fusion was 64.61 J/g in the control, which changed to 68.98, 52.70, 49.71 and 72.47 J/g in the treated T1, T2, T3, and T4 samples respectively. However, the melting temperature did not show any considerable change in the treated selenium samples as compared to the control. The FT-IR spectra showed the absorption peak at 526 and 461 cm-1, which corresponding to metal oxide bonding vibration in the control and treated selenium powder respectively. Hence, overall data suggest that, the biofield energy treatment considerably altered the physical and thermal properties of selenium powder. Therefore, biofield energy treatment could make selenium even more useful nutrient in human body.

Abstract Introduction:: In areas with severe selenium deficiency higher incidence of thyroiditis has been reported due to a decreased activity of selenium-dependent glutathione peroxidase enzyme within thyroid cells. Aims and Objective:: To study the effect of selenium supplementation in patients with autoimmune thyroid disease. Materials and Methods:: This is a blinded placebo-controlled prospective study done in 60 patients with autoimmune thyroid disease (as defined by an anti-thyroid peroxidase antibody (TPOAb) level more than 150 IU/ml) irrespective of the baseline thyroid status. Patients with overt hyperthyroidism who are on antithyroid drugs, patients on any other medication, which may alter the immunity status of the patients, and pregnant patients were excluded from the study. Patients were randomized into two age and TPOAb-matched groups; 30 patients received 200 μg of sodium selenite/day, orally, for 3 months, and 30 patients received placebo. All hypothyroid patients were given l-thyroxine replacement. Results:: Of 30 patients in the selenium treated group, 6 patients were overtly hypothyroid, 15 were subclinical hypothyroid, 6 were euthyroid, and 3 were subclinical hyperthyroid. The mean TPOAb concentration decreased significantly by 49.5% (P < 0.013) in the selenium treated group versus 10.1% (P < 0.95) in the placebo-treated group. Conclusion:: Selenium substitution has a significant impact on inflammatory activity in thyroid-specific autoimmune disease. It would be of interest to determine whether early treatment with selenium in patients with newly developed autoimmune thyroiditis may delay or even prevent the natural course of these diseases.

Materials and Methods:: To determine if leishmaniasis has effects on trace elements status, they were determined by atomic absorption spectrometry (AAS) in patients (n = 155, 95 CL and 60 VL) and control group (n = 100). Results:: Our findings indicate that there is a significant difference in the values of Se and Zn between control and patient groups (P < 0.0001 and P < 0.001, respectively). Se and Zn levels were 3.65 ± 0.88 and 67.24 ± 18.76 μg/dL in the leishmaniasis patients, and these values were observed to be statistically lower compared to the control groups (11.10 ± 2.37 and 119.61 ± 26.18 μg/dL, respectively). Meanwhile, no significant difference in status of Cu was found between the cases (110.55 ± 29.25 μg/dL) and healthy subjects (91.42 ± 27.54 μg/dL) (P > 0.05). When the patients were divided into two groups, there appeared to be a significant decrease (P < 0.001) in Se concentration for VL patients (2.57 ± 0.64 μg/dL) compared with CL patients (4.33 ± 1.06 μg/dL). Conclusion:: Based on these results, serum Se and Zn levels could be a useful marker for the pathophysiology of leishmaniasis.

Selenized yeast has also recently been shown to reduce lung cancer incidence and mortality in a population of skin cancer patients. Smokers and survivors of early stage lung and head and neck cancers have had a long period of promotion by carcinogenic agents on the bronchial epithelium resulting in morphologic and molecular alterations. We hypothesize that these morphologic and molecular alterations can be detected and modulated by chemopreventive agents. We have proposed a Phase lla chemoprevention trial evaluating five different dose levels of selenium administered daily for 3 months in subjects at high risk for lung cancer with bronchoscopically documented dysplasia. Subjects will be entered to determine the effect of dose on the modulation of biomarkers in response to selenium supplementation as well as to measure selenium levels and modulation of glutathione peroxidase as a measure of drug effect. In addition to morphology, the surrogate endpoint biomarkers to be examined include apoptosis, p53 expression, K-ras mutation analysis, p16 methylation, and upregulation of hnRNP A2/B1. Successful completion of this study will support selenium supplementation as potentially beneficial therapy in preventing the progression of lung carcinogenesis as well as identify surrogate endpoint markers that appear to be modulated by selenium supplementation.

The briefing discusses a project to develop an antibiotic that will selectively kill Yersinia pestis without killing other bacteria, while using a killing mechanism that Y. pestis can not develop a resistance to. This requires a target mechanism to bind to a specific protein on the Y. pestis, leading the researchers to look at peptides. Peptides are less expensive, more stable and easier to deliver than antibodies. The project examined the hypothesis that selenium labeled peptides and selenium labeled bacterial viruses (phage) can be produced that can selectively bind to the surface of a pathogenic bacteria and inactivate them through the generation of superoxide radicals on their surface.
