Abstract Background: We describe the genome of the western painted turtle, Chrysemys picta bellii, one of the most widespread, abundant, and well-studied turtles. We place the genome into a comparative evolutionary context, and focus on genomic features associated with tooth loss, immune function, longevity, sex differentiation and determination, and the species' physiological capacities to withstand extreme anoxia and tissue freezing. Results: Our phylogenetic analyses confirm that turtles are the sister group to living archosaurs, and demonstrate an extraordinarily slow rate of sequence evolution in the painted turtle. The ability of the painted turtle to withstand complete anoxia and partial freezing appears to be associated with common vertebrate gene networks, and we identify candidate genes for future functional analyses. Tooth loss shares a common pattern of pseudogenization and degradation of tooth-specific genes with birds, although the rate of accumulation of mutations is much slower in the painted turtle. Genes associated with sex differentiation generally reflect phylogeny rather than convergence in sex determination functionality. Among gene families that demonstrate exceptional expansions or show signatures of strong natural selection, immune function and musculoskeletal patterning genes are consistently over-represented. Conclusions: Our comparative genomic analyses indicate that common vertebrate regulatory networks, some of which have analogs in human diseases, are often involved in the western painted turtle's extraordinary physiological capacities. As these regulatory pathways are analyzed at the functional level, the painted turtle may offer important insights into the management of a number of human health disorders.

Chocolatier Pat Godfrey has boxes of freshly made Easter chocolates in her Plymouth shop, yet it's just a fraction of what her small business will produce between now and Easter. Godfrey's small, family-run business, Painted Turtle Chocolatier, opened in 2008 during the height of the recession, yet this artisan chocolatier business has stayed afloat thanks to a loyal customer base and strong holiday sales. "There's essentially four big seasons, it's Christmas, Valentine's, Easter, and then Thanksgiving," said Godfrey. "Every time there's a holiday, you pretty much work non--stop for about 3 weeks." Between now and Easter, Godfrey and her husband Doug, an internal medicine physician by day who helps make chocolate Easter bunnies and truffles on weekends, will produce 1,000 Easter bunnies and thousands of truffles. "At Valentine's we had 35 flavors of truffles, and they were all fresh, made within one week," said Godfrey. The majority of the chocolate at Painted Turtle comes in from Belgium. It arrives in either ten pound blocks or pellets before it's melted down in a tempering unit. In the shop's production room, a steady stream of chocolate flows out of the tempering units. Doug Godfrey patiently fills acryllic molds in the shape of bunnies with liquid milk chocolate. "You have to make sure you get chocolate down into the ears because the ears are the most susceptible part of the bunny," said Doug Godfrey. "They can snap off, we like to leave that to the kids to snap off the ears." After the molds are filled, they're refridgerated for 30 minutes before the chocolate is ready to be taken out. Godfrey says each chocolate bunny takes about 35 minutes to create from start to finish. Truffles are a more labor intensive, three day process. For Pat Godrey, who is a registered dietician and food scientist, becoming a chocolatier has meant creating a business people can't help but love. "As soon as they walk in, they smile. Chocolate makes people happy," said Godfrey. While controlling for potentially confounding variables, we compared relative abundance of painted turtles (Chrysemys picta) in 20 ponds in Eastern Ontario, 10 as close as possible to high traffic roads (Road sites) and 10 as far as possible from any major roads (No Road sites). There was no significant effect of roads on painted turtle relative abundance. Furthermore, our data do not support other predictions of the road mortality hypothesis; we observed neither a higher relative frequency of males to females at Road sites than at No Road sites, nor a lower average body size of turtles at Road than at No Road sites. We speculate that, although roads can cause substantial adult mortality in turtles, other factors, such as release from predation on adults and/or nests close to roads counter the negative effect of road mortality in some populations. We suggest that road mitigation for painted turtles can be limited to locations where turtles are forced to migrate across high traffic roads due, for example, to destruction of local nesting habitat or seasonal drying of ponds. This conclusion should not be extrapolated to other species of turtles, where road mortality could have a larger population-level effect than on painted turtles.
