This study is examining how best to convey the idea that maternal vaccination is an injunctive norm. MTurk participants will be screened to select 1000 women who are pregnant or trying to become pregnant. Eligible participants will be assigned to one of four experimental conditions: 1) Baby-focused messages with social norms; 2) Baby-focused messages without social norms; 3) Society-focused messages with social norms; 4) Society-focused messages without social norms. They will read one message about the importance of flu vaccination and one message about the importance of measles vaccination. For each disease message, participants will rate the message itself on several dimensions, their attitudes towards vaccination for this disease, how normal vaccination is for this disease, and their intentions to receive vaccination for this disease (or provide this vaccination to their child, in the case of measles). All participants will complete demographic questions.

Parents of students entering seventh grade are reminded that new immunization requirements announced earlier this year require students to have one Tdap (Tetanus-diphtheria-attenuated pertussis) and one meningococcal (MCV4) vaccination. Factors Affecting Uptake of Routine Immunization Among Children Age 12-23 Months in District Rahimyar Khan, Punjab, Pakistan , TIJFS

This is the process. Bacteria such as Haemophilus influenzae stick to the respiratory mucosa Bacteria divide and colonise the mucosa (forming a microbiome) Bacteria are coughed up in sputum Sputum is swallowed, therefore bacteria are swallowed Most bacteria are simply digested in the stomach A small percentage make it through to the small intestine These bacteria are taken up by immune lymphoid tissue in the wall of the small intestine called Peyer’s patches Peyer’s patches recognise these bacteria as foreign Peyer’s patches produce a form of T helper cell to combat the infection These newly formed T cells migrate in the blood to the lungs, then into the lung mucosa Here the new T helper cells ‘help’ the activation of immune responses such as the formation of more neutrophils These neutrophils are phagocytic, they eat any foreign material, such as any bacteria and any virus. They are not fussy eaters; they eat any antigen. The person now enjoys ‘learned innate immunity’ As bacteria and viruses are eaten and internally digested in the upper part of the lungs, they are dead, so cannot migrate down to infect the alveoli, the gaseous exchange structures. As the alveoli are not infected, they do not become inflamed, therefore they do not fill up with inflammatory fluid leading to a consolidated pneumonia As these thin-walled alveoli are not infected, the viruses and bacteria are not present, so do not migrate into the blood leading to systemic illness. Therefore, in short, the person stays well As the T cells are very powerful stimulants of immunity and inflammation, we do not want them hanging around for a long time, therefore they are short lived in the lungs However, the neutrophils they have stimulated self-perpetuate over a longer period of time (probably about 10 months) This means the upper airways are patrolled and protected for months after an episode of T cell stimulation As the upper airways are kept clear of viruses and bacteria the lower airways are protected from descending infection Therefore, infections such as influenza or covid may present as mild or even asymptomatic Such a protected person is therefore able to ‘compartmentalise’ the infection in the upper airways, preventing it from descending to the lower alveoli Physiologically, this system is protective but imperfect, for example a lot of the bacteria are digested in the stomach This means some people will be well protected from severe disease after covid or influenza infection, while others are not Giving an optimised strain of Haemophilus influenzae as an Immunobiotic can enhance the efficiency of this natural physiological system The administered Haemophilus influenzae is killed, so cannot reproduce to cause disease The administered Haemophilus influenzae is enteric coated, so it will be protected as it passes through the stomach, meaning it will reach the small intestine in large numbers This will provide lots of stimulation to the Peyer’s patches This is not a vaccination, it is an augmentation of a completely natural physiological system A tablet could probably be taken before every winter as an OTC (over the counter medication) Millions of acute exacerbations (going to the lungs) could be prevented The tablet would cost very little as an OTC, it’s just dead bugs Free textbook download, https://drjohncampbell.co.uk/

Immune responses are generally analyzed at the end of protocols including at least 2 administrations, but rarely after a prime. To investigate this point, we compared B and T cell responses in mice after one and two intrarectal immunizations with 2/6 rotavirus-like particles (2/6-VLP) and LT-R192G. After a boost, we found, an unexpected lower B cell expansion measured by flow cytometry, despite a secondary antibody response. We then analyzed CD4+CD25+Foxp3+ regulatory T cells (Tregs) and CD4+CD25+Foxp3− helper T cells after in vitro (re)stimulation of mesenteric lymph node cells with the antigen (2/6-VLP), the adjuvant (LT-R192G) or both. 2/6-VLP did not activate CD4+CD25+Foxp3− nor Foxp3+ T cells from non-immunized and 2/6-VLP immunized mice, whereas they did activate both subsets from mice immunized with 2/6-VLP in the presence of adjuvant. LT-R192G dramatically decreased CD4+CD25+Foxp3+ T cells from non-immunized and 2/6-VLP immunized mice but not from mice immunized with 2/6-VLP and adjuvant. Moreover, in this case, LT-R192G increased Foxp3 expression on CD4+CD25+Foxp3+ cells, suggesting specific Treg activation during the recall. Finally, when both 2/6-VLP and LT-R192G were used for restimulation, LT-R192G clearly suppressed both 2/6-VLP-specific CD4+CD25+Foxp3− and Foxp3+ T cells. All together, these results suggest that LT-R192G exerts different effects on CD4+CD25+Foxp3+ T cells, depending on a first or a second contact. The unexpected immunomodulation observed during the recall should be considered in designing vaccination protocols.

In year one of the current project, experiments have been performed to: (1) Examine the "normal" progression of SIV infection and disease using standardized conditions and the various assays proposed for examination of animals receiving DNA vaccines (including virus load, cellular and humoral immune responses, and cytokine profiles). Standardization of our SIV macaque animal model system was performed in preparation for genetic immunization versus SIV immunization. Assays for viral load, antiviral and cellular immune responses and cytokine levels (to SIV and nonspecific antigens) were applied to four SIV infected and two uninfected rhesus macaques. Considerable innate individual animal variation was observed in immune responsiveness to SIV and other antigens, as was corresponding variation in ability to control SIV infection. Our laboratory assays also revealed, somewhat unexpectedly, problems which now largely been resolved. (2) Develop novel expression vectors intended to improve levels of expression of SIV proteins from DNA injection in mice and primates. A number of novel expression vectors have been designed and tested in cell culture and in animals. These expression vector constructs were found to be very active relative to previously available constructions, and are now being adapted for the expression of a variety of SIV envelope proteins.

We cloned into a pVax expression vector the secreted and the truncated (no peptide leader sequence) versions of the human prostate acid phosphatase (H-PAP-T), the human prostate- specific antigen (H-PSA-T) and the rat analogue of the human PSMA (R-PSMA-S). A stable transfectant with H-PSMA and H-PSA of the AT3B1 cell line was obtained. The following plasmids were produced and purified under GLP-conditions using the Qiagen technology: H-PSMA-T, R-PSMA-T, H-PSA, H-PSA-T, H-PAP-T and RPSMA-S. Preliminary studies using the Copenhagen rat tumor prostate model showed uniform tumor development in rats that were injected subcutaneously with 100 000 AT3B-lPSMA,PSA cells. Using a commercially available transfection device from Amaxa, we could transfect differentiated dendritic cells with 20-40% efficiency. Such transfected dendritic cells stimulated in vitro autologous T cells to PSMA. T cells cytotoxicity was then tested against tumor cells or peptide-pulsed T2 target cells. Both H-PSMA-T DCs and S-PSMA DCs generated antigen-specific cytotoxic T cell responses. The immune response was restricted towards one of four PSMA derived epitopes when priming and boosting was performed with S-PSMA. In contrast, T-PSMA transfected DCs primed T cells towards several PSMA derived epitopes. Subsequent repeated boosting with transfected DCs restricted the immune response to a single immunodominant epitope.

This report provides updated information concerning the potential adverse events associated with vaccination for hepatitis B, poliomyelitis, measles, mumps, diphtheria, tetanus, and pertussis. This information incorporates findings from a series of recent literature reviews, conducted by an expert committee at the Institute of Medicine (lOM), of all evidence regarding the possible adverse consequences of vaccines administered to children. This report contains modifications to the previously published recommendations of the Advisory Committee on Immunization Practices (ACIP) and is based on an ACIP review of the lOM findings and new research on vaccine safety. In addition, this report incorporates information contained in the Recommendations of the Advisory Committee on Immunization Practices: Use of Vaccines and Immune Globulins in Persons with Altered Immunocompetence (MMWR 1993 ;42 ?No. RR-4!) and the General Recommendations on Immunization: Recommendations of the Advisory Committee on Immunization Practices (ACIP) (MMWR l994;43 ?No. RR-l !). Major changes to the previous recommendations are highlighted within the text, and specific information concerning the following vaccines and the possible adverse events associated with their administration are included: hepatitis B vaccine and anaphylaxis; measles vaccine and a) thrombocytopenia and b) possible risk for death resulting from anaphylaxis or disseminated disease in immunocompromised persons; diphtheria and tetanus toxoids and pertussis vaccine (DTP) and chronic encephalopathy; and tetanus-toxoid-containing vaccines and a) Guillain-Barre syndrome, b) brachial neuritis, and c) possible risk for death resulting from anaphylaxis. These modifications will be incorporated into more comprehensive ACIP recommendations for each vaccine when such statements are revised.

A review of the history of vaccine therapy for infectious diseases is presented. The concept originated when Auzias-Turenne introduced syphylitic vaccination as a treatment for syphilis in Paris in the mid-1800s; his clinical studies probably influenced Pastuer's successful rabies postexposure vaccine trials. Robert Koch in Berlin in the 1890s observed that innoculation of tuberlculin into patients with tuberculosis induced an inflammatory response in affected tissues, and advocated tuberculin therapy . Sir Almoth Wright in the early 20th century devised methods to measure changes in serum opsonizing activity in response to therapeutic innoculations with microbe-derived vaccines. Advances in antigen production and in molecular immunology now permit new tactics to probe, analyse and selectively alter in vivo human immune responses to infectious microbes. This article is from PLoS ONE , volume 9 . Abstract Haemophilus influenzae type b (Hib) is now recognized as an important pathogen in Asia. To evaluate disease susceptibility, and as a marker of Hib transmission before routine immunization was introduced in Kathmandu, 71 participants aged 7 months–77 years were recruited and 15 cord blood samples were collected for analysis of anti-polyribosylribitol phosphate antibody levels by enzyme-linked immunosorbent assay. Only 20% of children under 5 years old had levels considered protective (>0.15 µg/ml), rising to 83% of 15–54 year-olds. Prior to introduction of Hib vaccine in Kathmandu, the majority of young children were susceptible to disease.

This article is from PLoS ONE , volume 8 . Abstract Clostridium difficile is a pathogen which is responsible for diarrhea and colitis, particularly after treatment with antibiotics. Clinical signs are mainly due to two toxins, TcdA and TcdB. However, the first step of pathogenesis is the colonization process. We evaluated C. difficile surface proteins as vaccine antigens in the hamster model to prevent intestinal colonization. This vaccination induced a partial protection of hamsters against death after a C. difficile challenge. A proteomic analysis of animal sera allowed us to identify proteins which could be responsible for the protection observed. Among these proteins, we identified the GroEL heat shock protein. To confirm the role of the specific GroEL antibodies in the delayed C. difficile colonization of hamsters, we performed an immunization assay in a mouse model. After intranasal immunization with the recombinant protein GroEL, we observed a lower C. difficile intestinal colonization in the immunized group as compared to the control group.

Abstract Background: Virtually all low- and middle-income countries are dependent on the World Health Organization’s Expanded Program on Immunization for delivery of vaccines to children. The Expanded Program on Immunization delivers routine immunization services from health facilities free of charge. Understanding interventions for improving immunization coverage remains key in achieving universal childhood immunization. Methods: We will conduct a systematic review that aims to assess the effectiveness of the full range of potential interventions to improve routine immunization coverage in children in low- and middle-income countries. We will include intervention studies, as well as observational studies. We will search the Cochrane Database of Systematic Reviews (CDSR), Cochrane Central Register of Controlled Trials (CENTRAL), MEDLINE, EMBASE, electronic databases for eligible studies published by 31 August 2013. At least two authors will independently screen search outputs, select studies, extract data and assess the risk of bias (using separate criteria for interventions and observational studies); resolving any disagreements by discussion and consensus. The use of logic models and the Cochrane Complexity Matrix will be explored in order to better understand and contextualize studies. We will express the result of each study as a risk ratio with its corresponding 95% confidence intervals for dichotomous data, or mean difference with its standard deviation for continuous data. We will conduct meta-analysis for the same type of participants, interventions, study designs, and outcome measures where homogeneity of data allows. Use of harvest plots may be explored as an alternative. Heterogeneity will be assessed using the χ2 test of heterogeneity, and quantified using the I2 statistic. This protocol has not been registered with PROSPERO. Discussion: This review will allow us to document evidence across a broad range of intervention types for improving routine immunization coverage in children and also distinguish between those that are well supported by evidence (to direct policy recommendations) and those that are not well supported (to direct research agenda).

Several studies have revealed that an efficient HCV vaccine model should be able to induce both humoral and cell mediated immune responses targeting the conserved regions of the virus to circumvent the immune escape mutants. The beta chemokine Macrophage Inflammatory Protein 3-beta (MIP-3beta) is a key modulator of dendritic cells (DCs) and T-cells interaction, functions during immune response induction and is secreted specifically by cells in the lymphoid tissues. Objectives:: In the present study, we questioned whether co-administration of MIP-3beta gene could enhance the immune responses to HCV core in DNA vaccination. Materials and Methods:: Expression and biological activity of MIP-3beta expressing plasmid were evaluated by ELISA and transwell migration assays, respectively. HCV core DNA vaccine ± plasmid expressing MIP-3beta were electroporated subcutaneously to the front foot pads of BALB/c mice on days 0 and 14, and HCV core protein booster was applied to all core-DNA-vaccine received mice on the day 28. Both cell mediated immunity (proliferation, IFN-γ and IL-4 cytokine release, IFN-γ ELISpot and cytotoxic Granzyme B release assays) and humoral immune responses (total IgG and IgG2a/IgG1 subtyping) were evaluated ten days after final immunization. Results:: Mice covaccinated with MIP-3beta elicited an enhanced Th1 biased systemic immune response as evidenced by higher IFN-γ/IL-4 and anti-core IgG2a/IgG1 ratio, lymphoproliferation, strong cytolytic GrzB release and enhanced population of IFN-γ producing immunocytes. Likewise, the humoral immune response assumed as the total anti-core IgG level was augmented by MIP-3beta co-delivery. Conclusions:: These results exhibited the immuno potentiator effects of MIP-3beta plasmid when coadministrated with the HCV core DNA vaccine. Complimentary studies integrating MIP-3beta as a genetic adjuvant in HCV-core-DNA vaccination models are warranted.

Abstract Introduction: Since the comprehensive peace agreement was signed in 2005, institutionalization of immunization services in South Sudan remained a priority. Routine administrative reporting systems were established and showed that national coverage rates for DTP-3 rose from 20% in 2002 to 80% in 2011. This survey was conducted as part of an overall review of progress in implementation of the first EPI Multi-Year Plan for South Sudan 2007-2011. This report provides maiden community coverage estimates for immunization. Methods: A cross sectional community survey was conducted between January and May 2012. Ten cluster surveys were conducted to generate state-specific coverage estimates. The WHO 30x7 cluster sampling method was employed. Data was collected using pre-tested, interviewer guided, structured questionnaires through house to house visits. Results: The fully immunized children were 7.3%. Coverage for specific antigens were; BCG (28.3%), DTP-1(25.9%), DTP-3 (22.0%), Measles (16.8%). The drop-out rate between the first and third doses of DTP was 21.3%. Immunization coverage estimates based on card and history were higher, at 45.7% for DTP-3, 45.8% for MCV and 32.2% for full immunization. Majority of immunizations (80.8%) were received at health facilities compared to community service points (19.2%). The major reason for missed immunizations was inadequate information (41.1%). Conclusion: The proportion of card-verified, fully vaccinated among children aged 12-23 months is very low at 7.3%. Future efforts to improve vaccination quality and coverage should prioritize training of vaccinators and program communication to levels equivalent or higher than investments in EPI cold chain systems since 2007.

The recently widowed Adela Gereth, a lover of beauty and passionate collector of fine objects, strikes up a friendship with the young Fleda Vetch, when both of them find themselves guests in the tasteless house of Brigstock family. Mrs. Gereth fears that her son Owen, an honorable but somewhat unimaginative young man, may take up with one of the Brigstock girls, and indeed he presently announces his engagement to Mona, the eldest daughter. That means that Mrs. Gereth will have to leave Poynton, the beautiful house that she and her husband filled with the furniture, china, tapestries, and other objects that they lovingly collected over the years. It is not so much possessiveness that drives Mrs. Gereth to want to maintain control over them (or so she claims, at any rate), but rather the sense that she will have failed if Mona, understanding and appreciating nothing of what Poynton contains, should become Owen's wife and take charge. The story and its developing conflicts are seen largely from the point of view of Fleda Vetch, the young woman who, her moral and aesthetic sensibilities tuned perhaps as finely as any of James's protagonists, finds herself caught in the middle. James provides a popularizing outline of his view of philosophical pragmatism while making highly rhetorical and entertaining lashes towards rationalism and other competing schools of thought. James is especially concerned with the pragmatic view of truth. True beliefs should be defined as, according to James, beliefs that can successfully assist people in their everyday life. This is claimed to not be relativism. That reality exists is argued to be a fact true beyond the human subject. James argues, nevertheless, that people select which parts of reality are made relevant and how they are understood to relate to each other. Charles Sanders Peirce, widely considered to be the founder of pragmatism, eventually chose to separate himself intellectually from James, renaming his own theory to ‘pragmaticism’.
