1Isolation And Identification Of Antifungal Compounds From Bacillus Subtilis C9 Inhibiting The Growth Of Plant Pathogenic Fungi.

This article is from Mycobiology , volume 40 . Abstract Antagonistic microorganisms against Rhizoctonia solani were isolated and their antifungal activities were investigated. Two hundred sixteen bacterial isolates were isolated from various soil samples and 19 isolates were found to antagonize the selected plant pathogenic fungi with varying degrees. Among them, isolate C9 was selected as an antagonistic microorganism with potential for use in further studies. Treatment with the selected isolate C9 resulted in significantly reduced incidence of stem-segment colonization by R. solani AG2-2(IV) in Zoysia grass and enhanced growth of grass. Through its biochemical, physiological, and 16S rDNA characteristics, the selected bacterium was identified as Bacillus subtilis subsp. subtilis. Mannitol (1%) and soytone (1%) were found to be the best carbon and nitrogen sources, respectively, for use in antibiotic production. An antibiotic compound, designated as DG4, was separated and purified from ethyl acetate extract of the culture broth of isolate C9. On the basis of spectral data, including proton nuclear magneric resonance (1H NMR), carbon nuclear magneric resonance (13C NMR), and mass analyses, its chemical structure was established as a stereoisomer of acetylbutanediol. Application of the ethyl acetate extract of isolate C9 to several plant pathogens resulted in dose-dependent inhibition. Treatment with the purified compound (an isomer of acetylbuanediol) resulted in significantly inhibited growth of tested pathogens. The cell free culture supernatant of isolate C9 showed a chitinase effect on chitin medium. Results from the present study demonstrated the significant potential of the purified compound from isolate C9 for use as a biocontrol agent as well as a plant growth promoter with the ability to trigger induced systemic resistance of plants.

An antifungal and antibacterial antibiotic produced by the ectotrophic mycorrhizal fungus Leucopaxillus cerealis var. piceina has been identified as diatretyne nitrile, using biological and spectrophotometric techniques. Maximum production of this antibiotic occurred during the rapid and incipient autolysis growth phases in liquid medium. During continued autolysis the nitrile was reduced to diatretyne amide and diatretyne 3, which are antibacterial but not antifungal substances. Concentrations causing minimum inhibition (20%) to germination of zoospores of Phytophthora cinnamomi were 50-70 parts/billion (ppb), with total (100%) inhibition at 2 parts/million (ppm) . Approximately half of the inhibited zoospores germinate upon removal (dialysis) of diatretyne nitrile between these concentrations, indicating a fungistatic effect. Zoospores failed to germinate after removal of higher concentrations, indicating that these concentrations were lethal. Concentrations causing minimum inhibition (20%) to growth from mycelial discs of P. cinnamomi were detected at 0.5 ppm, with 100% inhibition at 9 ppm. Soil bacteria were inhibited between 0.5 and 2.5 ppm. The fungus produced approximately 12 ppm of diatretyne nitrile in liquid culture after 50 days' growth at 15 C. Diatretynes were produced in a variety of aseptic organic substrates to which sucrose or malt extract was added. Diatretyne nitrile did not inhibit germination of aseptic shortleaf pine seeds exposed for 1 or 2 hr to concentrations up to 40 ppm, but it was phytotoxic to aseptic seedlings at concentrations exceeding 10 ppm.

The 18th century book A Treatise on Modern Falconry is still one of the most quoted books in today's falconry community. <br><br>The preface was possibly written by a Rev. A. Willis, ridicules "The Origin and Progress of Language," a publication shortly before his writing this book. The introduction to this book gives a historical account of the development of falconry from its beginnings long before the 9th century in the middle East to its apparent decline at the invention of the hunting gun. It also includes interesting facts about the practice of falconry in Persia and Hindostan, where falconry became (and in some parts of the Middle East still is) a sport and pastime of the upper classes. <br><br>In the second and main part, the book talks about the falconer and his instruments as well as how to treat and train his birds. Robert Campbell (1829-84) was a Jamaican-born printer, journalist, and teacher who, along with Martin Robison Delany (1812-85), made up the Niger Valley Exploring Party of 1859-60 (Library of Congress), sharing his impressions & observations as a first time visitor to areas of modern day Nigeria. This work is a forerunner of "Back to Africa" movements later spearheaded by Jamaican compatriot, Marcus Garvey Jr (who was born two years after Campbell's death). - Summary by Faith Abiola-Ellison
