Within the framework of the project 'Acoustic Microsensors' (A93KL448) the possibilities of a biosensor system based on an immunological reaction on a piezoelectric crystal sensor are investigated at the TNO Prins Maurits Laboratory (TNO-PML). The results described are a continuation of previous reported work in which the toxin Staphylococcal Enterotoxin B (SEB) was detected in a competition assay with a preincubation period of at least 20 minutes and a detection limit of 100 ng/ml. A new biochemical interface based on a flexible carboxylated dextran matrix, to which antibodies are bound, was developed. With this biosensor system SEB could be detected directly without a preincubation step at a lowest concentration of approximately 7 ng/ml. The biosensor can be easily regenerated by a one-minute injection of 0.1 M HCl.
