4Response Inhibition In Children With Fragile X Syndrome And Autism

Autism spectrum disorder (ASD) (hereafter: autism) is a neurodevelopmental condition that is characterized by social difficulties and restrictive and repetitive behaviors. Fragile X syndrome (FXS) is the most common single-gene cause of autism (Kidd et al., 2014; Lozano et al., 2016; Stone et al., 2024). Children with FXS and children with autism often display difficulties with executive functioning (Schmitt et al., 2007). One particular domain on which children with FXS (Sullivan et al., 2007; Tonnsen, 2016) and children with autism (Hlavatá et al., 2018) are known to have issues is response inhibition. In this study we aim to increase our understanding of the neurocognitive phenotype of children and adolescents with FXS or autism by comparing them with a typically developing (TD) group on their response inhibition capabilities. We will compare our groups on response inhibition using behavioral data from an inhibition task and the performance on a subtask of the NEPSY-II. In addition, we will explore group differences in pupil sizes during our inhibition task. It has been shown that pupil size increases during tasks that require cognitive control (e.g. a task requiring response inhibition), but also decrease as a task becomes too demanding and induces cognitive fatigue (Mahot, 2018). Therefore, comparing pupillometric data between our study groups can give us insights on the cognitive load demands of response inhibition children and adolescents with FXS or autism.

Aversion to eye contact is a common behavior of individuals diagnosed with Fragile X syndrome (FXS); however, no studies to date have attempted to increase eye-contact duration in these individuals. In this study, we employed a percentile reinforcement schedule with and without overcorrection to shape eye-contact duration of 6 boys with FXS. Results showed that although aversion to eye contact is often thought to be unamenable to change in FXS, it can be shaped in some individuals using percentile schedules either alone or in combination with overcorrection. 8Group I Metabotropic Glutamate Receptor Mediated Dynamic Immune Dysfunction In Children With Fragile X Syndrome.

This article is from Journal of Neuroinflammation , volume 11 . Abstract Background: Fragile X syndrome (FXS) is the leading cause of inheritable intellectual disability in male children, and is predominantly caused by a single gene mutation resulting in expanded trinucleotide CGG-repeats within the 5’ untranslated region of the fragile X mental retardation (FMR1) gene. Reports have suggested the presence of immune dysregulation in FXS with evidence of altered plasma cytokine levels; however, no studies have directly assessed functional cellular immune responses in children with FXS. In order to ascertain if immune dysregulation is present in children with FXS, dynamic cellular responses to immune stimulation were examined. Methods: Peripheral blood mononuclear cells (PBMC) were from male children with FXS (n = 27) and from male aged-matched typically developing (TD) controls (n = 8). PBMC were cultured for 48 hours in media alone or with lipopolysaccharides (LPS; 1 μg/mL) to stimulate the innate immune response or with phytohemagglutinin (PHA; 8 μg/mL) to stimulate the adaptive T-cell response. Additionally, the group I mGluR agonist, DHPG, was added to cultures to ascertain the role of mGluR signaling in the immune response in subject with FXS. Supernatants were harvested and cytokine levels were assessed using Luminex multiplexing technology. Results: Children with FXS displayed similar innate immune response following challenge with LPS alone when compared with TD controls; however, when LPS was added in the presence of a group I mGluR agonist, DHPG, increased immune response were observed in children with FXS for a number of pro-inflammatory cytokines including IL-6 (P = 0.02), and IL-12p40 (P
