Abstract Gas exchangers fundamentally form by branching morphogenesis (BM), a mechanistically profoundly complex process which derives from coherent expression and regulation of multiple genes that direct cell-to-cell interactions, differentiation, and movements by signaling of various molecular morphogenetic cues at specific times and particular places in the developing organ. Coordinated expression of growth-instructing factors determines sizes and sites where bifurcation occurs, by how much a part elongates before it divides, and the angle at which branching occurs. BM is essentially induced by dualities of factors where through feedback- or feed forward loops agonists/antagonists are activated or repressed. The intricate transactions between the development orchestrating molecular factors determine the ultimate phenotype. From the primeval time when the transformation of unicellular organisms to multicellular ones occurred by systematic accretion of cells, BM has been perpetually conserved. Canonical signalling, transcriptional pathways, and other instructive molecular factors are commonly employed within and across species, tissues, and stages of development. While much still remain to be elucidated and some of what has been reported corroborated and reconciled with rest of existing data, notable progress has in recent times been made in understanding the mechanism of BM. By identifying and characterizing the morphogenetic drivers, and markers and their regulatory dynamics, the elemental underpinnings of BM have been more precisely explained. Broadening these insights will allow more effective diagnostic and therapeutic interventions of developmental abnormalities and pathologies in pre- and postnatal lungs. Conservation of the molecular factors which are involved in the development of the lung (and other branched organs) is a classic example of nature’s astuteness in economically utilizing finite resources. Once purposefully formed, well-tested and tried ways and means are adopted, preserved, and widely used to engineer the most optimal phenotypes. The material and time costs of developing utterly new instruments and routines with every drastic biological change (e.g. adaptation and speciation) are circumvented. This should assure the best possible structures and therefore functions, ensuring survival and evolutionary success.

2Cell-Based Multi-Parametric Model Of Cleft Progression During Submandibular Salivary Gland Branching Morphogenesis.

Abstract Cleft formation during submandibular salivary gland branching morphogenesis is the critical step initiating the growth and development of the complex adult organ. Previous experimental studies indicated requirements for several epithelial cellular processes, such as proliferation, migration, cell-cell adhesion, cell-extracellular matrix (matrix) adhesion, and cellular contraction in cleft formation; however, the relative contribution of each of these processes is not fully understood since it is not possible to experimentally manipulate each factor independently. We present here a comprehensive analysis of several cellular parameters regulating cleft progression during branching morphogenesis in the epithelial tissue of an early embryonic salivary gland at a local scale using an on lattice Monte-Carlo simulation model, the Glazier-Graner-Hogeweg model. We utilized measurements from time-lapse images of mouse submandibular gland organ explants to construct a temporally and spatially relevant cell-based 2D model. Our model simulates the effect of cellular proliferation, actomyosin contractility, cell-cell and cell-matrix adhesions on cleft progression, and it was used to test specific hypotheses regarding the function of these parameters in branching morphogenesis. We use innovative features capturing several aspects of cleft morphology and quantitatively analyze clefts formed during functional modification of the cellular parameters. Our simulations predict that a low epithelial mitosis rate and moderate level of actomyosin contractility in the cleft cells promote cleft progression. Raising or lowering levels of contractility and mitosis rate resulted in non-progressive clefts. We also show that lowered cell-cell adhesion in the cleft region and increased cleft cell-matrix adhesions are required for cleft progression. Using a classifier-based analysis, the relative importance of these four contributing cellular factors for effective cleft progression was determined as follows: cleft cell contractility, cleft region cell-cell adhesion strength, epithelial cell mitosis rate, and cell-matrix adhesion strength.

The purpose of this study is to determine the role of a transcriptional factor, Msx2, in regulating initial induction of mammary anlagen and subsequent branching events that may require cooperative assistance by a few other molecules. Due to the time limitation of the funding period, we focused our attention on one key event that Msx2 gene may be an essential player, i.e., the initial inductive event to form the mammary gland. We took advantage the fact that we have Msx2 transgenic animals in that the Msx2 transgene is controlled by the Msx2 promoter. We have shown that this promoter is active in the mesenchyme of mammary anlagen around the time mammary gland is form in the mouse. We hypothesized that Msx2 transgenic animals should be able to rescue the mammary gland development in the Msx2 null mutants. As predicted, we were able to rescue mammary gland development in the Msx2 null mutants when the transgene is present. In addition, we also found that the transgene enhanced branching at the expense of extension. We also noticed that in a small percentage of Msx2 null mutants, mammary gland did development; however, mammary glands in these animals showed fewer branches. Taken together these results, we concluded that the Msx2 gene is essential for the initial induction event, and in addition it regulates numbers of branches.

Abstract Proper lung functioning requires not only a correct structure of the conducting airway tree, but also the simultaneous development of smooth muscles and vasculature. Lung branching morphogenesis is strongly stereotyped and involves the recursive use of only three modes of branching. We have previously shown that the experimentally described interactions between Fibroblast growth factor (FGF)10, Sonic hedgehog (SHH) and Patched (Ptc) can give rise to a Turing mechanism that not only reproduces the experimentally observed wildtype branching pattern but also, in part counterintuitive, patterns in mutant mice. Here we show that, even though many proteins affect smooth muscle formation and the expression of Vegfa, an inducer of blood vessel formation, it is sufficient to add FGF9 to the FGF10/SHH/Ptc module to successfully predict simultaneously the emergence of smooth muscles in the clefts between growing lung buds, and Vegfa expression in the distal sub-epithelial mesenchyme. Our model reproduces the phenotype of both wildtype and relevant mutant mice, as well as the results of most culture conditions described in the literature.

5Mechanisms Of Side Branching And Tip Splitting In A Model Of Branching Morphogenesis.

This article is from PLoS ONE , volume 9 . Abstract Recent experimental work in lung morphogenesis has described an elegant pattern of branching phenomena. Two primary forms of branching have been identified: side branching and tip splitting. In our previous study of lung branching morphogenesis, we used a 4 variable partial differential equation (PDE), due to Meinhardt, as our mathematical model to describe the reaction and diffusion of morphogens creating those branched patterns. By altering key parameters in the model, we were able to reproduce all the branching styles and the switch between branching modes. Here, we attempt to explain the branching phenomena described above, as growing out of two fundamental instabilities, one in the longitudinal (growth) direction and the other in the transverse direction. We begin by decoupling the original branching process into two semi-independent sub-processes, 1) a classic activator/inhibitor system along the growing stalk, and 2) the spatial growth of the stalk. We then reduced the full branching model into an activator/inhibitor model that embeds growth of the stalk as a controllable parameter, to explore the mechanisms that determine different branching patterns. We found that, in this model, 1) side branching results from a pattern-formation instability of the activator/inhibitor subsystem in the longitudinal direction. This instability is far from equilibrium, requiring a large inhomogeneity in the initial conditions. It successively creates periodic activator peaks along the growing stalk, each of which later on migrates out and forms a side branch; 2) tip splitting is due to a Turing-style instability along the transversal direction, that creates the spatial splitting of the activator peak into 2 simultaneously-formed peaks at the growing tip, the occurrence of which requires the widening of the growing stalk. Tip splitting is abolished when transversal stalk widening is prevented; 3) when both instabilities are satisfied, tip bifurcation occurs together with side branching.

3Autobiography of a Super-Tramp

The Autobiography of a Super-Tramp is an autobiography published in 1908 by the Welsh poet and writer W. H. Davies (1871–1940). A large part of the book's subject matter describes the way of life of the tramp in the United Kingdom, Canada and the United States in the final decade of the 19th century. George Bernard Shaw had become interested in Davies, a literary unknown at the time, and had agreed to write a preface for the book, largely through the concerted efforts of his wife Charlotte. Shaw was also instrumental in keeping the unusual title of the book, of which Davies himself was unsure, and which later proved to be controversial with some reviewers. The book was the third published by Davies, having been preceded by The Soul's Destroyer (1905) and New Poems (1907). The 1920 edition of the book concludes with five poems selected by Davies from The Soul's Destroyer. The book was written in the space of six weeks, "a great achievement for a first book by a man with the minimum of education." ( Wikipedia (edited by Expatriate))

W.H.Davies was born in Newport, South Wales. He had a difficult childhood and spent several years in the US as a tramp or hobo. He spent one winter moving between jails in Michigan under the ‘boodle’ system; an illicit system where a tramp could make an arrangement to be housed in a prison without being an actual prisoner. He later lost his right foot below the knee after a failed attempt to jump a freight train in Ontario on a journey to the Klondike to try to make his fortune.<br> On his return to the UK he continued to live rough or in doss-houses for some time, before eventually settling down with his wife Helen and moving on to a quiet and happy life.<br> Poetry was his staple throughout his life and he was eventually recognised for his talents.<br> Much of his poetry reflects on themes of hardship and the natural world.<br> Summary by Craig Franklin
